摘 要:目的探讨乙醇对氨茶碱游离药物浓度的影响。方法采用中空纤维离心超滤(HFCF-UF)前处理技术结合高效液相色谱(HPLC)法对氨茶碱游离药物浓度进行测定,并比较乙醇加入前后游离药物浓度的变化。结果加入乙醇后氨茶碱游离药物浓度增加,且血浆乙醇浓度与游离药物浓度呈线性相关关系。结论加入乙醇可能会影响血浆中的氨茶碱与血浆蛋白的结合,游离药物浓度增加,易出现毒副作用及毒性反应。Objective To investigate the effect of alcohol on free drug concentration of aminophylline. Methods Hollow fiber centrifugal ultrafiltration( HFCF-UF) combined with HPLC was adopted to determine the free drug concentration of aminophylline. The difference in free concentration before and after alcohol adding into the plasma was further compared to explore the impact on it of different alcohol concentrations. Results The free drug concentration of aminophylline increased with the addition of alcohol,meanwhile,there was a linear relationship between it and the alcohol concentration in plasma. Conclusion The addition of ethanol may affect the binding rate of aminophylline and plasma proteins in plasma,increase the free drug concentration,and prone to side effects and toxicity.
