摘 要:目的探讨通心络(TXL)对同型半胱氨酸(Hcy)诱导大鼠心肌微血管内皮细胞线粒体功能损伤的保护作用及机制。方法用Hcy建立大鼠心肌微血管内皮细胞损伤模型,分为对照组、Hcy组、TXL-L组、TXL-M组、TXL-H组。分别检测各组细胞线粒体活性、线粒体膜电位(MMP)和细胞内活性氧(ROS)水平。结果与对照组相比,模型组细胞线粒体活性和MMP降低,细胞内ROS水平升高(P<0.05,P<0.01);与模型组相比,TXL各浓度组均能不同程度地改善上述指标变化(P<0.05,P<0.01)。结论 TXL能够改善Hcy诱导的大鼠心肌微血管内皮细胞线粒体功能障碍,其作用机制可能与降低细胞内ROS水平有关。Objective To observe the protective effects of Tongxinluo( TXL) on homocysteine( Hcy)-induced rat cardiac microvascular endothelial cells( RCMECs) mitochondrial dysfunction,and to determine the mechanism behind its protection. Methods Hcy was used to develop model,the cells were divided into control,Hcy,Tongxinluo low,middle and high groups. The cell mitochondrial activity,mitochondrial membrane potential( MMP),content of adenosine triphosphate( ATP),reactive oxygen species( ROS) level and protein expression of Cytochrome C( Cyt C) were observed.Results Compared with those of Control group,cell mitochondrial activity,MMP and ATP concentration were decreased in Hcy group,ROS production and protein expression of Cyt C were increased in Hcy group( P0.05,P0.01).Compared with those of Hcy group,the index of different changes of TXL every dose group were improved,and the high concentration of tongxinluo group was the best( P0.05,P0.01). Conclusions TXL could improve Hcy-induced dysfunction of mitochondria in rat cardiac microvascular endothelial cells,and plays a protective role by reducing ROS level in the cells.
