HPLC法同时测定蒙药复方述达格-4中四个黄酮类成分的含量

摘　　要：建立同时测定蒙药复方述达格-4中山奈酚、高良姜素、山奈素、高良姜素-3-甲醚含量的方法.采用HPLC法,色谱柱为AgilentC18色谱柱(4.6mm×250mm,5μm),流动相乙腈-0.2%乙酸水溶液(梯度洗脱),流速0.6mL·min-1,检测波长254nm,柱温30℃,进样量为10μL.得出山奈酚、高良姜素、山奈素和高良姜素-3-甲醚线性范围分别为0.00067-0.00670μg(r=0.9999)、0.5240-5.2400μg(r=0.9999)、0.1484-1.4840μg(r=0.9999)、0.0996-0.9960μg(r=0.9999);精密度、稳定性、重复性试验的RSD<2%;平均加样回收率分别为96.79%(RSD=1.70%,n=6)、97.38%(RSD=1.59%,n=6)、100.96%(RSD=1.49%,n=6)、97.01%(RSD=1.50%,n=6).结果表明该方法操作简便、稳定、重复性好,可用于蒙药复方述达格-4中4个黄酮成分含量的测定.本研究可为蒙药复方述达格-4资源开发利用提供理论依据.To establish a method for simultaneous contents determination of kaempferol,galan gin,kaempferide and galangin-3-methylether in Mongolian medicine Shudage-4. HPLC was performed on the column of Aglient C18 with mobile phase of acetonitrile-0. 2% glacial acetic acid （gradient elution）at a flow rate of 0.6mL/min, detection wavelength was 254nm, column tempera- ture was 30℃, and injection volume was 10gL. The linear range was 0. 00067 -- 0. 00670gg for kaempferol （r=0. 9999）. 0. 5240- 5. 2400μg for galangin （r=0. 9999）. 0. 1484- 1. 4840t.tg for kaempferide （r=0. 9999）.and 0. 0996-0. 9960μg for galangin-3-methylether （r= 0. 9999） ;RSD of precision,stability and reproducibility tests were lower than 2%;average recoveries were 96. 79~ （RSD=l. 70~,n=6）.97. 38% （RSD= l. 59% ,n=6）.100.960//0（RSD=l. 49%,n=6）.97.01% （RSD=1. 50%,n=6）. The method is simple,stable,reproducible and can be used for the contents determination of four flavonoids in Mongolian medicine Shudage-4. The study can provide theoreti- cal basis for developing and using Mongolian medicine Shudage-4.
