摘  要:目的建立以高效液相色谱法同时测定月华丸(胶囊)及其含药血清中地黄苷A、麦冬皂苷D、原百部碱三种有效成分的方法,对月华丸(胶囊)及其含药血清进行质量控制。方法以地黄苷A、麦冬皂苷D和原百部碱为对照,采用Hypersil BDS C18色谱柱(250 mm×4.6 mm,5um)。其中地黄苷A流动相∶乙腈∶水(6∶94),检测波长:203nm;麦冬皂苷D的流动相:乙腈∶水(48∶52),检测波长:208nm;原百部碱流动相:乙睛∶0.25%甲酸铵(25∶75),检测波长:305 nm。流速:1ml/min;柱温:35℃,进样量:20μl。结果按峰面积法计算,月华丸(胶囊)及其血中移行成分的地黄苷A、麦冬皂苷D和原百部碱的含量分别为0.276、0.161、0.508 mg/g和0.104、0.098、0.141mg/g。结论该方法可以用于月华丸(胶囊)及其入血成分的定性鉴定、定量测定和质量控制。Objective A method for simultaneous determination of Rehmaionoside A,ophiopogonin D,protostemonine is implemented to control the quality of Yuehua pill( capsule) and its composition into the blood via HPLC. Methods With rehmaionoside A,ophiopogonin D and protostemonine as control,this research uses Hypersil BDS C18column( 250 mm × 4. 6,mm,5um),in this circumstance,ophiopogonin D: mobile phase acetonitrile: water( 48: 52),the detection wavelength: 208nm; rehmaionoside A mobile phase acetonitrile: water( 6: 94),the detection wavelength: 203nm; protostemonine mobile phase: acetonitrile:0. 25% formic acid ammonium( 25: 75),the detection wavelength: 305 nm; flow rate: 1ml/min,column temperature: 35 degrees centigrade,sample size: 20 μL experiment. Results Through required calculation the confirmation of determination of rehmannioside A,ophiopogonin D,protostemonine were 0. 104,0. 098,0. 141mg/g in Yuehua pill( capsule) and its composition into the blood. Conclusion The contents of this method by Yuehua pill( capsule) into blood components as indexes could provide the scientific basis for better controlling the quality.
