电针干预对吗啡耐受形成中小鼠的生物节律及其相关钟基因Per 1和Per 2表达的影响

摘  要:目的观察电针干预对吗啡受形成中小鼠的生物节律及其相关钟基因Per1和Per2表达的影响。方法将小鼠24只,随机分为空白对照组(n=8),耐受模型组(n=8),电针干预组(n=8)。耐受模型组、电针干预组皮下注射盐酸吗啡注射液以建立吗啡耐受模型。电针干预组每日上午注射吗啡后开始用电针行双侧"肾俞"穴(BL 23)电针治疗。各组在第1、3、5、7d行小鼠光热甩尾潜伏期测定。第7 d在测定痛阈后处死小鼠取视交叉上核(Suprachiasmatic nucleus,SCN)组织,用荧光定量PCR(RT-PCR)方法检测小鼠SCN中Per1、Per2基因的表达变化。结果 (1)痛阈:耐受模型组和电针干预组在第1、3、5d注射吗啡后,测得痛阈值明显升高,与其初痛阈相比差异明显;电针干预组第7d痛阈与初痛阈相比有差异。(2)自发性昼夜活动节律:耐受模型组喝电针干预组与空白对照组相比,小鼠周期、总活动期、振幅结果均有差异:空白对照组与耐受模型组和电针干预组相比小鼠实验前后振幅差值有差异,耐受模型组与电针治疗组相比具有差异。(3)Per1、Per2基因在SCN中的表达水平:与空白对照组相比,耐受模型组的相对表达量下调;电针干预组与耐受模型组相比较结果又明显差异。结论电针干预组方法可以通过上调吗啡耐受模型小鼠的节律相关基因Per1、Per2的表达量来减缓吗啡耐受小鼠痛阈值的下降趋势,延缓耐受的形成并纠正振幅降低为表现的生物节律紊乱。Objective To observe electro-acupuncture regulate the circadian rhythms and its expression of clock gene period1 and period 2 on morphine tolerance mice. Methods 24 male mice were randomly divided into 3 groups,Control group( n = 8),Model group( n = 8),Electro-acupuncture group( n = 8). The Model group and Electro-acupuncture group were built the morphine tolerance models by injecting 10 mg/kg morphine hydrochloride injection. the Electro-acupuncture group began to receive treatment by using the electro-acupuncture at bilateral " Shenshu" point( BL 23) for 7 days consecutively. We measured the tail flick latency as an index of pain threshold at the 1st,3rd,5th and 7th day. Animals were sacrificed respectively in the morning at the 7th day and collected the samples of SCN of mice and tested the the expression of Per1 gene and Per2 gene in SCN of mice by RT-PCR method. Results(1)Pain threshold: Pain threshold at the 1st,3rd and 5th day of both model group and electro-acupuncture group compared with their initial values,has significant differences( P〈0. 05). Pain threshold at the 7th day of electro-acupuncture group compared with its initial valus,still has significant differences( P〈0. 05).(2)The circadian rhythm: The Electro-acupuncture group and Model group had significant differences in amplitude and activity compared with the Control group( P〈0. 05). There also had significant differences between Model and Electro-acupuncture group in amplitude( P〈0. 05). Conclusion Electro-acupuncture can somehow inhibit morphine tolerance formation through regulation of genes Penes Period 1 and Period 2.
