5种商品化试剂提取病毒RNA的比较研究

摘　　要：选择商品化的3种细胞裂解液(RNAiso Plus、TRNzol Plus和RNAOUT)和2种DNA/RNA共提试剂盒(RNA-DNA双提试剂盒、快速DNA/RNA共提试剂盒),分别提取猪繁殖与呼吸综合征病毒(PRRSV)JXA1-R株、猪瘟病毒(CSFV)HCLV株和乙型脑炎病毒(JEV)SA-14-14-2株的RNA,通过RNA直接定量和Real-time PCR间接评价了抽提的RNA的质量。RNA直接检测结果表明,RNAiso Plus、TRNzol Plus和RNAOUT 3种细胞裂解液抽提获得的RNA浓度相对较高;Real-time PCR间接检测结果表明,快速DNA/RNA共提试剂盒组RNA相对水平最高,RNAiso Plus和TRNzol Plus 2种细胞裂解液组其次。研究表明,RNAiso Plus、TRNzol Plus和RNAOUT 3种试剂可用于临床RNA病毒检测以及定量分析。Three commercially available cell lysates（ RNAiso Plus,TRNzol Plus and RNAOUT） and two DNA/RNA co-kits（ LC-DNA double-kit,rapid DNA/RNA co-kit） were selected to extract three animal RNA viruses（ PRRSV,CSFV and JEV）. The RNA quality was evaluated indirectly by agarose gel electrophoresis and Real-time PCR. The Real-time PCR result showed that the RNA relative levels of the rapid DNA/RNA co-kit group was highest,and the two cell lysates groups（ RNAiso Plus,TRNzol Plus） followed. For RNA concentration,the levels of the Three commercially available cell lysates were better than the rapid DNA/RNA co-kit and LC-DNA double-kit. It suggests that the above three reagents can be used for clinical RNA virus detection and quantitative analysis.
