摘  要:目的探讨麦冬总皂苷和麦冬多糖对香烟烟雾提取物(CSE)刺激的人支气管上皮细胞(16HBE细胞)生长增殖的影响,以及炎症相关因子的表达。方法体外培养16HBE细胞,采用MTT法检测CSE、麦冬总皂苷、麦冬多糖的细胞毒性,根据实验结果确定受试物浓度。麦冬总皂苷和麦冬多糖均设置高、中、低3个剂量,孵育16HBE细胞24 h后给予CSE刺激,用MTT法测定细胞的生长情况,用ELISA法检测细胞上清液中表皮生长因子(EGF)、转化生长因子-β_1(TGF-β_1)、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、白细胞介素-10(IL-10)的表达。结果与模型组相比,麦冬总皂苷、麦冬多糖能提高16HBE细胞的相对生长率,上调细胞上清液中EGF、TGF-β_1的表达,使上清液中TNF-α、IL-6含量明显降低,IL-10含量明显升高,呈剂量依赖性。结论麦冬总皂苷、麦冬多糖能促进16HBE细胞的生长增殖,促进生长因子释放,抑制炎症反应。Objective To determine the effect and mechanism of ophiopogonin and ophiopogon polysaccharides on the proliferation and inflammatory factor of human bronchial epithelial cells(16HBE) damaged by cigarette smoke extract(CSE). Methods The 16 human bronchial epithelial cells were cultured in vitro. The cytotoxicity of CSE, ophiopogonin and ophiopogon polysaccharides was measured by MTT assay to determine the final concentrations. We treated 16 HBE cells with high, medium and low concentration of ophiopogonin and ophiopogon polysaccharides for 24 h and damaged with CSE. MTT assay was used to detect the growth of cells. The levels of EGF, TGF-β_1, TNF-α, IL-6, and IL-10 were also measured. Results Compared with the model group, the relative growth rate of 16 HBE cells treated with ophiopogonin and ophiopogon polysaccharides was increased. Ophiopogonin and ophiopogon polysaccharides reduced TNF-α and IL-6, increased IL-10 levels obviously in a dose-dependent manner. Ophiopogonin and ophiopogon polysaccharides improved EGF and TGF-β_1 levels. Conclusion Ophiopogonin and ophiopogon polysaccharide have remarkable effect on the proliferation of 16 HBE cells induced by CSE. The mechanism may be associated with promoting the proliferation of 16 HBE cells, inhibiting the inflammatory reaction and elevating the levels of growth factors.
