摘  要:目的建立超高效液相-四极杆-飞行时间质谱(UPLC-EIS-Q-TOF-MS)法分析灯台叶颗粒活性成分。方法色谱条件:Agilent Extend C_(18)(2.1 mm×100 mm,1.8μm),流动相为0.1%甲酸胺水(A)-乙腈(B),梯度洗脱,体积流量为0.4 mL·min^(-1),质谱使用ESI离子源正离子模式下采集数据,并运用Agilent Masshunter Qualitative Analysis工作站中的分子特征提取(MFE)功能,根据提供化合物的准确相对分子质量、质谱碎片离子信息,并结合参考文献等进行成分鉴定。结果从灯台叶颗粒中鉴定出27种化学成分,黄酮及其苷类7个,生物碱类7个,萜类10个,氨基酸类3个。芹菜素、齐墩果酸羧甲酯、齐墩果酸羧乙酯、β-香树脂醇、苯丙氨酸、环(酪氨酸-酪氨酸)、N-乙酰-L-脯氨酸7个化合物均为首次从灯台草制剂中被发现并报道。结论 UPLC-ESIQ-TOF-MS技术能够快速、准确地鉴定灯台叶颗粒中的化学成分,为其质量控制和临床应用提供了理论依据。Objective To analyse and identify the chemical constituents from Dengtaiye particles with UPLCESIQ-TOF-MS. Methods The analysis was performed on an Agilent Extend C_(18)(2.1 mm×100 mm, 1.8 μm). The mobile phase consisted of 0.1% ammonium formate(A)-acetonitrile(B), and gradient elution was used, with the flow rate of 0.4 mL·min^(-1). Mass specromentry was applied for the quanlitative analysis under positive ion mode and ESI ion source. Data was analyzed by Agilent Masshunter Qualitative Analysis and MFE. According to the provided accurate relative molecular mass, the mass spectrometry in compound ions, information, and references, we identified the chemical constituents. Results Totally 27 chemical components were identified in Dengtaiye particles, involving 7 flavonoids and its glycosides, 7 alkaloids, 10 terpenoids, 3 amino acids. Seven chemical constituents such as apigenin, oleanolic acid carboxy methylester, oleanolic acid carboxy ethyl ester, β-amyrin, phenylalanine, ring(tyrosine-tyrosine), and N-acetyl-L-proline were first reported in Dengtaiye and its preparations. Conclusion UPLC-ESIQ-TOF-MS method can identify the main chemical constituents from Dengtaiye particles rapidly and accurately, which provides a new strategy for its quality control as well as a reference for its efficacy.
