摘  要:目的:心肌的缺血再灌注氧化应激反应是细胞损伤的主要原因,对氧磷酶(PON2)是一种在体内广泛分布的细胞内抗氧化物,本实验拟探讨PON2是否参与心肌缺血再灌注损伤过程。方法:本实验收集风湿病外科手术患者体外循环前后的少许心房组织,用QPCR方法进行心肌标本的PON2的mRNA水平的检测,并分析其与临床数据的相关性。结果:风湿病患者心房组织内有PON2的mRNA的表达,而且缺血再灌注前后有表达水平的变化,男性患者PON2的mRNA的表达下调,而女性患者的PON2的mRNA的表达上调,PON2的表达水平的变化与缺血时间、再灌注时间无明显相关性。结论:提示PON2参与了风湿病患者外科手术缺血再灌注的过程,而且这种变化在不同性别之间有明显的区别,初步提示PON2可能为不同患者的心功能预后提供一个判断依据。Objective: oxidative stress caused by myocardial ischemia reperfusion were the main cause of cardiac cell damage. Paraoxonase( Paraoxonase 2,PON2) is an intracellular antioxidant widely distributed in vivo. This experiment is to study whether PON2 is involved in myocardial ischemia reperfusion injury. Methods: a few atrial tissues were collected before and after cardiopulmonary bypass in patients with rheumatic diseases. The mRNA level of PON2 was detected by QPCRmethod,and the correlation between the mRNA level of PON2 and the clinical data was analyzed. Results: The expression of PON2 mRNA was detected in atrial tissues of patients with rheumatic diseases. And the changes of PON2 mRNA level were found before and after ischemia reperfusion. In male patients the level of PON2 mRNA was down-regulated,inversely the level of PON2 mRNA was up-regulated in female patients before and after ischemia reperfusion. No obvious correlation was found between the mRNA level of PON2 and the clinical data. Conclusion: Theseresult suggest that PON2 is involved in the process of ischemia reperfusion in rheumatism surgical patients. The obvious difference of PON2 mRNA level in different gender suggests that PON2 may be an important factor for judging the prognosis of cardiac function in different patients.
