益气活血方对凝血酶诱导内皮细胞促血栓因子表达的影响

摘　　要：目的:观察益气活血方对凝血酶诱导内皮细胞表达促血栓因子纤溶酶原激活物抑制剂-1(plasminogen activator inhibitor-1,PAI-1)及组织因子(tissue factor,TF)的影响。方法:体外培养内皮细胞,在含有凝血酶(4 U·m L^(-1))的培养基中加入梯度质量浓度的益气活血方(0~5.0 g·L^(-1)),作用12 h后MTT法检测细胞活力,确定益气活血方对内皮细胞无毒性的最大浓度;之后将细胞分为4组进行实验,分别为空白对照组、凝血酶组、益气活血方低剂量组(0.25 g·L^(-1))、益气活血方高剂量组(1.25 g·L^(-1)),加入益气活血方预处理2 h后加入凝血酶刺激10 h,Western blot检测PAI-1、TF的表达量。结果:由MTT检测结果得到益气活血方对内皮细胞无毒性的最大浓度为1.25 g·L^(-1),并将此浓度作为益气活血方高剂量组。Western blot结果显示益气活血方低剂量组(0.25 g·L^(-1))、高剂量组(1.25 g·L-1)均可抑制凝血酶诱导的PAI-1、TF表达(P<0.05或0.01),且呈一定的量效关系。结论:益气活血方可有效抑制凝血酶诱导内皮细胞促血栓因子PAI-1、TF的表达,对内皮细胞有潜在保护作用。Objective： To observe the effect of Yiqi Huoxue Decoction on the expression of pro-thrombotic factor plasminogen activator inhibitor-1（ PAI-1） and tissue factor（ TF） in endothelial cells induced by thrombin. Methods： Endothelial cell EA. hy926 was cultured in vitro in the culture medium with thrombin（ 4 U·m L-1）,as well as different concentration of Yiqi Huoxue Decoction（ 0-5 g·L-1） for 12 hrs. Cell viability of EA. hy926 was detected by using MTT essay to determine the maximal concentration of Yiqi Huoxue Decoction without toxicity on EA. hy926 cells. The cells were then divided into 4 groups,namely control,thrombin,Yiqi Huoxue Decoction low dosage,and Yiqi Huoxue Decoction high dosage group. After the cells were pretreated with Yiqi Huoxue Decoction for 2 hrs and the following stimulation of thrombin for 10 hrs,the expression of PAI-1 and TF was detected by using western blot essay. Results： By MTT,it was found that the maximum of Yiqi Huoxue Decoction without toxicity to endothelial cells was 1. 25 g·L-1,and this dosage was used as Yiqi Huoxue Decoction high dosage group. The results of western blot measurement suggested that both Yiqi Huoxue Decoction low dosage（ 0. 25 g·L-1） and Yiqi Huoxue Decoction high dosage（ 1. 25 g·L-1）inhibited the expression of PAI-1 and TF induced by thrombin in a dose-dependent manner（ P 〈 0. 05,P 〈 0. 01）. Conclusion：Yiqi Huoxue Decoction can effectively inhibit the expression of pro-thrombotic factor PAI-1 and TF in endothelial cells induced by thrombin,and it has a potential protective effect on endothelial cells.
