摘 要:目的:研究壮医方白花丹消蛊汤对食蟹猴肝纤维化的干预作用。方法:将12只食蟹猴随机分为4组,除空白对照组外,其余各组每周2次皮下注射0.5 mL/kg 40%四氯化碳(CCl4)橄榄油剂造模,同时辅以高脂饲料喂养,建立食蟹猴肝纤维化模型。造模6个月后,各组相应给药,白花丹消蛊汤低、高剂量组分别给予1.0、4.0 g(生药)/(kg·d)灌胃,空白组和模型组予等体积的生理盐水。给药8周后,静脉采集食蟹猴血液样本,检测各组食蟹猴血清中丙氨酸氨基转移酶(ALT)、门冬氨酸氨基转移酶(AST)、碱性磷酸酶(ALP)、总胆红素(T-BIL),酶联免疫法(ELISA)检测食蟹猴血清中血小板衍生生长因子(PDGF)和白介素1β(1L-β)水平,苏木素伊红(HE)染色和Masson染色观察肝纤维化程度。结果:白花丹消蛊汤低、高剂量组血清ALT、AST、ALP、T-BIL、PDGF水平均低于模型组;与模型组比较,白花丹消蛊汤低、高剂量组基质金属蛋白酶、免疫组化、肝组织纤维化程度明显改善。结论:壮医方白花丹消蛊汤具有抗肝纤维化作用。Objective: To observe the effect of Zhuang medicine prescription of Baihuadan Xiaogu Tang in hepatofibrosis on cynomolgus macaque. Methods: The 12 Cynomolgus macaques were randomly divided into 4 groups. All cynomolgus macaques, except the animal of normal group, were induced by injection of 40% carbon tetrachloride hypodermically twice a week at dose of 0.5 mL/kg, and at the same time animal were given rich fat and drug diet. Six months after the modeling, Baihuadan Xiaogu Tang low dose group and high dose group were givenl.0 and 4.0 g/(kg·d) by garage respectively; blank group and model group were given equal volume of saline. After 8 weeks all cynomolgus macaques were sacrificed. The enzyme's content of ALT, AST, ALP, T-BIL and factor's leve of PDGF, 1L-β from blood were determined. The hepatofibrosis were observed through the staining of HE and Masson. Results: Baihuadan Xiaogu Tang low dose group and high dose group showed lower content of ALT, AST, ALP, T-BIL and PDGF from blood than model group. Compared with the model group, matrix metalloproteinase, immunohistochemistry, liver fibrosis significantly improved in Baihuadan Xiaogu Tang low dose group and high dose group. Conclusions: The prescription of Baihuadan Xiaogu Tang is capable of anti- hepatofibrosis action.
