枳实消痞丸对大鼠上消化道CCK及CCK-A受体mRNA表达的影响

摘　　要：目的:探讨枳实消痞丸方对大鼠胃及十二指肠黏膜胆囊收缩素及其A受体基因表达的影响.方法:♂Wistar大鼠20只,分为对照及用药组,每组10只.分别以1.5mL生理盐水及枳实消痞丸方水煎制剂灌胃4wk.处死后取材,用RT-PCR法检测十二指肠黏膜CCKmRNA及胃底、胃窦肌肉组织CCK-A受体mRNA表达水平,并以同管所扩β-actin作为内参照进行半定量比较.结果:中药组大鼠十二指肠黏膜CCKmRNA及胃底CCK-A受体mRNA的表达水平明显较对照组降低(1.13±0.26vs0.67±0.10,P<0.001;7.30±2.61vs3.37±1.07,P<0.01),但胃窦部的CCK-A受体mRNA的表达水平明显增高(1.22±0.24vs3.25±1.11,P<0.001).结论:枳实消痞丸方可能通过对CCKmRNA及CCK-A受体mRNA表达水平的影响,从而起到对消化道动力的某些影响及临床上的一些治疗作用.AIM:To observe the influence of the Zhishi Xiaopi pill onthe gene expression of cholecystokinin (CCK) and cholecys-tokinin-A receptor (CCK-AR) mRNA in stomach and duode-num in rat.METHODS:After the rats were douched by TCM herbs ofZhishi Xiaopi recipe or normal saline for 4 weeks, the levelof CCK and CCK-AR mRNA in gastric and duodenal tissueswere analyzed by RT-PCR. The densities of the electrophore-sis pattern were scaned and quantitatively measured bythe Kodak digital science 1D program according to beta-actin mRNA.RESULTS:Compared to the control group, the levels ofCCK mRNA in duodenal mucosa and CCK-A receptor mRNAin gastric fundus were significantly decreased respec-tively (1.13±0.26 vs 0.67±0.10, P <0.001; 7.30±2.61 vs3.37±1.07, P <0.01) while that of CCK-A receptor mRNA ingastic antrum was significantly increased (1.22±0.24 vs 3.25±1.11, P <0.001) in rats of TCM group.CONCLUSION:TCM herbs of Zhishi Xiaopi recipe canregulate the mRNA expression of CCK and CCK-A receptorin rat, which may be related to the effect of gastric empty-ing promotion and indicates some possible advantages inclinical use.
