Gαi2在单侧输尿管梗阻大鼠肾组织中的表达及意义

摘　　要：目的探讨Gαi2在单侧输尿管梗阻(UUO)大鼠肾组织中的表达变化及意义。方法选用36只SD大鼠,随机分为UUO模型组(n=18)以及对照组(sham组,n=18)。分别于术后3、7、14d处死(每个时间点n=6),分成UUO3d组、UUO7d组和UUO14d组。取左肾组织留存。分别采用HE染色和Masson染色法观察每个时间点肾脏组织病理变化,用免疫组织化学法和Western blot法检测每个时间点肾脏组织中Gαi2和Ⅰ型胶原的表达。结果 HE和Masson染色结果显示,随着梗阻时间延长,大鼠肾组织损伤逐渐加重,胶原沉积逐渐增多。免疫组织化学染色结果显示随着梗阻时间延长,大鼠肾脏组织中的Gαi2和Ⅰ型胶原含量逐渐增多,UUO组各组之间以及与对照组相比差异显著(P<0.05)。Western blot结果显示,随着梗阻时间延长,大鼠肾脏组织中的Gαi2和Ⅰ型胶原表达逐渐增高,UUO组各组之间以及与对照组相比差异显著(P<0.05)。相关性分析结果显示,随着大鼠肾脏梗阻时间延长,Gαi2和Ⅰ型胶原表达均与UUO大鼠肾间质胶原沉积呈正相关,Gαi2与Ⅰ型胶原表达呈正相关。结论在单侧输尿管梗阻大鼠肾脏组织中Gαi2和Ⅰ型胶原表达呈时间依赖性,随着梗阻时间延长二者含量逐渐增多,Gαi2可能在肾间质纤维化的发生发展过程中起到重要作用。Objective To investigate the expression of Gαi2 in renal interstitial fibrosis rats caused by unilateral ureteral obstruction and its clinical significance.Methods Thirty-six SD rats were divided into sham-operation group（n=18）and UUO model group（n=18）randomly.Rats were sacrificed at 3,7,and 14 day after operation established for model group（n=6 for each day）,and then the left kidney of rats was obtained.HE and Masson staining were used to evaluate the degree of tubulointerstitial fibrosis in UUO group and sham-operation group at each time point.The expression of Gαi2 and collagenⅠwere detected by immunohistochemistry and Western blot.Results The degree of renal tubulointerstitial fibrosis and collagen deposition in UUO group was severely than that in sham groups.Immunohistochemistry and Western blot showed that the expression of Gαi2 and collagenⅠincreased with the prolongation of obstruction time and were significantly different at each time point as well as compared with those in the sham-operation group（P〈0.05）.There was positive correlation between collagen deposition and Gαi2,collagenⅠ expression,and the positive expression of Gαi2 was positively correlated with collagenⅠ.Conclusion The expression of Gαi2 increases gradually with the obstruction time in the renal tissue of unilateral ureteral obstruction rats and it may be involved in the pathophysiology of tubulointerstitial fibrosis as a key role.
