摘  要:肠道微生物在植食性昆虫与寄主植物的协同进化中发挥着重要的作用。采用基于16S rRNA基因的变性梯度凝胶电泳(DGGE)技术分析了3种植食性叶蜂(河曲丝叶蜂(Nematus hequensis)、落叶松叶蜂(Pristiphora erichsonii)和松阿扁叶蜂(Acantholyda posticalis))幼虫的肠道菌群,旨在比较取食不同寄主植物的叶蜂幼虫的肠道菌群特征。结果表明,3种叶蜂幼虫肠道细菌组成存在明显差异,其中取食阔叶树的河曲丝叶蜂幼虫肠道菌群明显不同于取食针叶的2种叶蜂,后两者的菌群比较相似。河曲丝叶蜂幼虫肠道细菌隶属于变形菌门、厚壁菌门和放线菌门,包括8个属,其中沙雷氏菌属(Serratia)和根瘤菌属(Rhizobium)为优势菌群;落叶松叶蜂幼虫肠道细菌隶属于变形菌门和厚壁菌门,包括6个属,其中肠杆菌属(Enterobacter)和假单胞菌属(Pseudomonas)为优势菌群;松阿扁叶蜂幼虫肠道细菌隶属于变形菌门、厚壁菌门和软壁菌门,包括7个属,其中沃尔巴克氏体属(Wolbachia)和假单胞菌属(Pseudomonas)为优势菌群。因此,3种叶蜂的肠道菌群与其各自取食的寄主植物具有密切的关系,该研究为进一步研究肠道微生物在植食性昆虫对寄主植物的选择中所起的作用提供了基础信息。Gut bacteria play an important role in coevolution between herbivorous insects and their host plants. In this paper the bacterial communities from larvae of three herbivorous sawfly species, Nematus he- quensis ,Pristiphora erichsonii and Acantholyda posticalis ,were profiled by using 16S rRNA PCR-denatu ring gradient gel electrophoresis (PCR-DGGE) technology, aiming to compare the characteristics of gut bacterial communities among sawflies feeding on different host trees. The bacterial communities of three sawfly larvae showed obvious difference, the gut bacterial community in N. hequensis which feed on the leaves of deciduous tree was clearly different from those in two sawflies (P. erichsonii and A. posticalis) feeding pine needles, both of which showed more similarity in gut bacterial community. Eight genera of Proteobacteria, Firmicutes and Actinobacteria were detected in N. hequensis, and Serratia and Rhizobium were the most abundant genera. Six genera of Proteobacteria and Firmicutes were identified in P. erichsonii that was dominated by Enterobacter and Pseudomonas. The bacterial flora of A. posticali was dominated byWolbachia and Pseudomonas, while other five genera of Proteobacteria, Firmicutes and Tenericutes were detected in low abundance. So the bacterial compositions in three sawflies species are closely related with their feeding host trees, and our results would provide basic information to further research the roles of gut microbes in host selection for herbivorous insects.
