摘  要:细胞体外3D培养技术更有利于细胞功能的表达和发挥,是细胞组织工程的重要研究内容。马鹿鹿茸间充质干细胞(MSCs)是一种新发现的成体干细胞,具有向软骨分化的能力。【目的】为了深入研究鹿茸MSCs细胞特性及软骨分化机制,【方法】利用水凝胶为支撑材料进行3D培养,与细胞平面培养方式对比,分析鹿茸MSCs的生长特性与变化。【结果】结果表明,3D培养组生长呈团状,形态呈梭型。两种培养法细胞生长曲线相似,培养至48 h后细胞进入对数生长期,平台期即培养144 h之后3D培养组细胞密度显著高于平面培养组(p<0.05);对两种方法培养312 h(13 d)的细胞进行PI染色,3D培养组细胞凋亡率(32.67±11.08%)低于平面培养组(37.18±10.21%),但两组间没有显著差异。Three- dimensional (3D) cell culture modelsis are an important research content in the cell tissue engineering which are numerous advantages in enabling cells to acquire a natural 3D phenotype, including increased cell proliferation, differentiation and function. To study Three -dimensional (3D) cell culture modets of Antler mesenchymal stem cells (MSCs), [ Objective] cell growing were measured with MrFr and Cells proliferation was detected with PI stainingon hydrogel scaffold. [ Results ] The results showed that the cell in 3D cell culture were spindle - shaped in appearance and with dense growth. The growth curves in 3D cell culture were similar to that in monolayer culture. And cells were in the logarithmic growth period from 48 hours. Compared with monolayer culture, the cells density in 3 D cell culture was significantly higher after 144h, ( p 〈 O. 05 ). By PI staining, the apoptosis ratio of MSCs in 3D - culture was 37.18 + 10.21% after 312 h,were lower than that in monotayer culture that was 32.67 + 11.08%, but no significant differences were found between those two groups.
