摘  要:目的:用大肠杆菌表达重组B型肉毒毒素受体结合区Hc抗原(BHc)作为亚单位疫苗,并研究其免疫原性。方法:构建pTIG-Trx-BHc原核表达载体,用大肠杆菌表达并纯化重组BHc抗原;免疫BALB/c小鼠以研究它的免疫原性。结果:大肠杆菌表达并纯化获得了重组BHc抗原,免疫小鼠后能刺激机体产生高效价的抗BHc抗体和保护性免疫反应。该重组BHc亚单位疫苗1μg剂量2次免疫小鼠即可产生对104LD50剂量B型肉毒毒素攻毒的完全保护,血清中和效价可达1.33 IU/m L。结论:大肠杆菌表达的重组BHc抗原具有良好的免疫原性,能够有效地保护小鼠抵抗B型肉毒毒素的攻击,该BHc抗原能够作为亚单位候选疫苗预防B型肉毒毒素中毒。Objective: The recombinant Hc domain antigen of botulinum neurotoxin serotype B(BoNT/B) was expressed in Escherichia coli as a subunit vaccine, and its immunogenicity was studied. Methods: The prokaryotic expression vector pTIG-Trx-BHc was constructed, recombinant antigen BHc was expressed in E.coli and purified. BALB/c mice were immunized to study its immunogenicity. Results: The recombinant BHc antigen protein was suc- cessfully expressed and purified from E.coli and could stimulate the body to produce high titers of anti-BHc anti- bodies, and induce protective immune responses in immunized mice. The recombinant BHc subunit vaccine provid- ed complete protection against challenge with 10^4 LD50 of BoNT/B only after 1 μg of two doses immunization, and serum neutralization titers reached 1.33 IU/mL. Conclusion: These findings suggest that the recombinant BHc antigen expressed in E.coli is efficacious in protecting mice against challenge with BoNT/B and that the recombinant BHc subunit vaccine may be a subunit candidate vaccine for prophylaxis against BoNT/B.
