摘  要:传染性法氏囊病病毒(IBDV)超强毒(vvIBDV)不适应鸡胚成纤维细胞培养,其分离鉴定通常采用直接接种无特定病原体(SPF)鸡的方法。然而该方法需要特殊设备,成本较高,较繁琐。研究旨在探讨SPF鸡胚作为IBDV分离工具的可行性,将IBDV阳性病料混悬液通过绒毛尿囊膜途径接种9~11日胚龄SPF鸡胚,并盲传三代成功分离获得vvIBDV(IBD16GX01株)。分离毒各代次序列一致,且与SPF鸡分离氨基酸同源性100%。结果表明,SPF鸡胚可用于vvIBDV的分离鉴定,为vvIBDV分子流行病学研究奠定基础。Very virulent infectious bursal disease virus (vvIBDV)can not adapt to chicken embryo fibroblast culture. Isolation of vvIBDV was usually performed by directly inoculation of specific-pathogen-free (SPF) chickens. However, this method needs special equipment, higher costs, and is not convenient. The aim of this study was to explore the feasibility of SPF chicken embryo as IBDV isolation tool. The mixed suspension of IBDV position bursa was inoculatedwith 9-to-ll-day-age SPF chicken embryo via the chorioallantoic membrane (CAM)pathway, and the vvlBDV strain (IBD16GXO1) was successfully isolated via three blind-passage. The sequence per generation of the isolated virus was consistent, and showed 100% homologous rate of amino acids with the virus isolated using SPF chicken. The results indicated that SPF chicken embryo could be used for the isolation of vvIBDV, which could lay the foundation for the vvlBDV molecular epidemiology study.
