摘  要:目的:讨论携抗ICAM-1的靶向超声造影剂在诊断动脉粥样硬化中的可行性。方法使用碳二亚胺法,在聚合物微泡表面修饰携抗ICAM-1,使这种微泡能够靶向定位于动脉粥样硬化斑块。结果:光学显微镜和扫描电镜结果指出微泡具有可控的微米级的粒径,荧光显微镜和流式细胞仪显示了靶向配体和微泡之间较高的结合率,体内超声成像实验证实了微泡在靶向超声成像的利用性。结论:制备的微泡具有大小均匀,粒径分布窄和抗体携带率高等特点。Objective The feasibility of targeted ultrasound contrast agents conjugated with anti-lCAM-I in diagnosing atherosclerosis was discussed. Methods A conjugation strategy was adopted using polymeric shelled MBs to enable localized targeting on atherosclerotic plaque by modifying the surface of these polymeric MBs with anti-ICAM-l. Results Light microscope and SEM studies indicated that the MBs with a controlled size distribution in the desired sub-micrometer range. Fluorescence microscopy and flow cytometry analysis showed high conjugation efficiency of targeting ligands to MBs. In vivo ultrasound targeted imaging results demonstrated the availability of conjugated MBs in targeted ultrasound imaging. Conclusions The polyrner MBS have many good characteristic, such as small, tight size distribution and high conjugation efficiency. These properties enable targeted MBs to identify atherosclerotic plaques in vivo and are useful in targeted ultrasound contrast-enhanced imaging.
