摘 要:目的研究北京市西城区食源性和临床感染性单核细胞增生李斯特菌(listeria monocytogenes,Lm)的脉冲场凝胶电泳(plused-field gel electrophoresis,PFGE)和多位点序列分型(multilocus sequence typing,MLST)分型特征,分析两种分型方法的应用价值。方法采用PFGE和MLST两种方法对2012-2015年50株食源性和临床感染性单核细胞增生李斯特菌进行分型研究。结果 50株单核细胞增生李斯特菌PFGE分型得到23种型别,GX6A16.CN0004为优势带型。MLST分析获得12个型别,ST9为优势型别。其中5组数据是ST型别只对应一种PFGE型别,3种PFGE型别同时存在食源性和临床感染性单核细胞增生李斯特菌,并且其ST型别进化关系比较远。结论北京市西城区不同来源单核细胞增生李斯特菌污染长期存在,出现致病型别,并且致病性单核细胞增生李斯特菌的遗传关系呈多元化。两种分子分型方法的结合,从基因水平上解释了单核细胞增生李斯特菌的变化趋势及流行特征。Objective To understand the molecular characteristics of food-borne and human-derived Listeria monocytogenes in Xicheng district of Beijing by using pulsed-field gel electrophoresis(PFGE)and multilocus sequence typing(MLST),and to assess the application value of PFGE and MLST. Methods 50 strains of food-borne and human-derived Listeria monocytogenes were collected from 2012 to 2015 and were genotyped by using PFGE and MLST. Results Out of 50 strains of Listeria monocytogenes,23 PFGE and 12 MLST genotypes were identified with the dominant type of GX6A16.CN0004 and ST9.Three PFGE types contained both food-borne and human-derived Listeria monocytogenes,but their ST types showed a little genetic correlation. Conclusions Listeria monocytogenes food contamination is not uncommon and sometimes pathogenic with diverse genetic relationship.The combination of two molecular typing methods explains the change trends and epidemiological characteristics of Listeria monocytogenes from the genetic level.
