摘 要:目的探讨丙卡特罗对呼吸道合胞病毒(RSV)感染正常人支气管上皮细胞(NHBE)分泌IL-8、IL-13和IL-25水平的影响。方法选择7例毛细支气管炎住院患儿为喘息组,以同期住院的7例腹股沟斜疝、隐睾择期手术患儿作为对照组,采集2组患者的外周血标本,提取外周血单个核细胞(PBMC),建立RSV感染NHBE与PBMC共培养的体外细胞模型,观察IL-8、IL-13和IL-25的表达水平,以及丙卡特罗干预后上述炎症因子的变化。结果免疫荧光技术鉴定RSV感染NHBE细胞模型建立成功。RSV感染前后,喘息组IL-8、IL-13及IL-25水平均高于对照组(P均<0.05)。喘息组中,RSV感染组IL-8、IL-13和IL-25水平高于对照组,丙卡特罗干预组IL-8、IL-13水平比RSV感染组有所下降(P均<0.05)、2组间IL-25水平比较差异无统计学意义(P>0.05)。结论 RSV感染后,NHBE的IL-8、IL-13、IL-25水平上调,可能触发和加重气道炎性反应,而丙卡特罗可能通过下调IL-8和IL-13水平发挥抗炎作用。Objective To investigate the effect of procaterol on the secretion of interleukin(IL)-8, IL-13 and IL-25 by normal human bronchial epithelial cells (NHBE) infected by respiratory syncytial virus (RSV) in vitro. Methods Seven children diagnosed with capillary bronchiolitis were assigned into the asthma group, and 7 counterparts with inguinal hernia or cryptorchidism scheduled to undergo surgery were allocated into the control group. Peripheral blood mononuclear cells (PBMCs) were isolated from the peripheral blood samples in two groups. The RSV-infected NHBE were co-cultured with PBMCs. The changes in the expression levels of IL-8, IL-13 and IL-25 before and after procaterol intervention were quantitatively measured. Results The RSV-infected NHBE model was successfully established that was validated by immunofluorescent staining. Before and after procaterol intervention, the expression levels of IL-8, IL-13 and IL-25 in the asthma group were significantly higher compared with those in the control group (all P&lt;0.05). In the NHBE-infected by RSV group, the levels of IL-8, IL-13 and IL-25 were significantly up-regulated than those in the asthma group (all P&lt;0.05), whereas the expression levels of IL-8 and IL-13 were considerably down-regulated in the procaterol intervention (both P&lt;0.05). The expression of IL-25 did not significantly differ from the NHBE-infected by RSV group and the procaterol intervetion group (P&gt;0.05). Conclusions After infected with RSV, the expression levels of IL-8, IL-13 and IL-25 in NHBE are up-regulated, which possibly triggers and aggravates the incidence of respiratory inflammatory response. Procaterol can suppress the inflammatory response probably by down-regulating the expression of IL-8 and IL-13.
