摘 要:为了研究植物Fes1蛋白C末端特有的约40个氨基酸序列的生物学功能,以拟南芥为实验材料,将C末端缺失的AtFes1A基因转化进atfes1a突变体中,研究AtFes1A蛋白C末端序列是否为功能必需的结构域。结果表明,C末端缺失的AtFes1A可以表达稳定的转录本和蛋白,但C末端缺失的AtFes1A无法修复atfes1a的热敏感性,并且转基因株系中的热激蛋白HSP70的表达水平也未恢复,因此植物Fes1的C末端是植物特有且功能必需的。To study the biological function of about 40 amino acid sequences specific to the C-terminal of the plant Fesl, taking Arabidopsis thaliana as the experimental material to introduce the C-terminal deletion of AtFeslA gene into the atfesla mutant to investigate whether the C-terminal sequence of AtFeslA protein was an indispensable functional domain. The results show that AtFeslA without C-terminal can express stable transcripts and proteins. But AtFeslA without C-terminal presents neither remedy in heat-sensitive defect of atfesla nor the recovery of cytosolic HSP70 level. These findings suggest that the C-terminal is plant-specific and essential for normally performing the function of Fesl in plants.
