摘 要:目的:研究天麻素对模拟高原缺氧大鼠心脏的保护作用。方法:2月龄雄性Wistar大鼠60只,按随机数字表法随机分为常氧模型组、缺氧模型组、红景天(阳性对照)组、天麻素(35、75、105mg/kg)组,每组10只。每日定时灌胃,连续7天。采用低压氧舱模拟海拔8000 m高原环境,将除常氧模型组外的5组大鼠置入实验舱,缺氧12h后观察大鼠心组织病理显微结构的改变,并测定心组织相关生化指标水平。结果:与常氧模型组相比,缺氧12h后,缺氧模型组大鼠心组织出现病理损伤,心组织中乳酸(LD)含量、乳酸脱氢酶(LDH)活性、丙二醛(MDA)含量、过氧化氢(H2O2)含量显著上升,谷胱甘肽过氧化物酶(GSH-Px)活力显著下降。与缺氧模型组相比,天麻素(35、75、105mg/kg)组心组织病理损伤较小;天麻素(35、105mg/kg)组LD含量显著下降且与常氧模型组无差异,天麻素75mg/kg组LD含量显著下降但与常氧模型组有差异;天麻素(35、75mg/kg)组LDH活性有下降趋势但无统计学意义仍高于正常,天麻素105mg/kg组LDH活性有显著下降与常氧模型组无差异;天麻素(35、75、105mg/kg)组MDA含量、H2O2含量均显著下降,GSH-Px活性显著上升,且均与常氧模型无差异。结论 :研究表明天麻素对模拟高原缺氧大鼠心脏具有保护作用。Objective: To study the effect of gastrodin on heart of rats under simulated high altitude hypoxia environment. Methods: Totally 60 two months old male Wistar rats were randomly divided into the control group,the hypoxia model group,rhodiola crenulata group,low-,mediumand high-doses of gastrodin( 35,70,105mg/kg) groups according to random number table,10 in each group. The intragastric administration to rats was continued for 7 days,once per day. The low pressure oxygen cabin was used as the simulated 8000 m altitude,five groups of rats were sent to the chamber of the cabin except the control group. Pathological changes of brain tissues were observed and related indexes of brain were detected after 12 h hypoxia. Results: Compared with the control group,the pathological damage was inflicted in the hypoxia group,contents of lactic acid( LD) 、malondialdehyde( MDA),hydrogen peroxide( H202) and the activity of lactic dehydrogenase( LDH) were significantly increased and the activity of glutathione peroxidase( GSH-Px) was significantly decreased in hypoxia group. Damages of heart tissue were alleviated in gastrodin( 35,75,105mg/kg) groups compared with the hypoxia group. LD was decreased in gastrodin( 35,105mg/kg)groups,with no statistical significance compared with the control group,and LD was significantly decreased in 75 mg/kg gastrodin group compared with the control group. LDH decreased in gastrodin groups,but the decrease was not significant compared with the control group. In gastrodin( 35,75,105mg/kg) groups,contents of MDA and H2O2 were significantly decreased,the activity of GSH-Px was significantly increased,and the difference were not obvious compared with the control group. However,effects in three groups of gastrodin were not dose dependent. Conclusion: There was an protective effect of gastrodin on heart of rats under simulated high altitude hypoxia environment.
