摘  要:目的:探讨黄精地龙提取液对老年痴呆大鼠脑内M1、M2-AchR受体的影响。方法:采用颈背部皮下注射1%D-半乳糖联合腹腔注射东莨菪碱复制大鼠老年痴呆模型,再用2、4、8g/kg·d量的黄精地龙提取液灌胃痴呆大鼠,运用Morris水迷宫、尼氏染色、免疫组化观察各大鼠大脑皮层第一躯体感觉区(S1Tr)和海马CA1与CA3区的阳性神经元数量;RT-q PCR、Western blot检测各大鼠大脑内的M1,M2-AchR mRNA及蛋白的表达。结果:2~8g/kg·d剂量范围内的黄精地龙提取液能能改善衰老痴呆大鼠的行为、学习记忆能力;能增加痴呆大鼠大脑S1Tr区和海马CA1及CA3区神经元数量;能升高痴呆大鼠大脑内M1-AchR mRNA和蛋白的表达水平,但对M2-AchR mRNA和蛋白表达水平无明显影响。结论:黄精地龙提取液能保护大脑认知学习记忆有关重要区的神经细胞,同时触发M1受体后效应,提高中枢Ach含量,进而升高老年痴呆大鼠中枢胆碱能系统活性。Objective: To observe the effects of Huangjing Dilong extracts on M1,m2-AchR expression in senile dementia rat's brain. Methods:After making the senile dementia models by nuchal subcutaneous injected 1% D-galactose and by intraperitoneal injected Scopolamine in the rats,then intragastric administration of 2,4,8g/kg. d Huangjing Dilong extracts,we tested the learning and memory abilities of the rats by morris water maze,investigated the positive neurons of hippocampal CA1 and CA3 area and S1 Tr detected by Nissl staining and immunohistochemistry methods; The M1,M2-AchR mRNA and proteins in the senile dementia rat's brain measured by RT-q PCR and Western blot respectively. Results: Huangjing Dilong extracts( 2 ~ 8g/kg. d) can significantly increased the learning and memory abilities and decreased the wrong behaviors of the senile dementia rats,can increase the number of neurons in hippocampal CA1 and CA3 area and S1 Tr area;Huangjing Dilong extracts can increase the expression of M1-AchR mRNA and protein in the brain of dementia rats,however,there was no significant effect on the expression of M2-AchR mRNA and protein in the brain of dementia rats. Conclusion: Huangjing Dilong extracts can protect more nerve cells which in the brain's cognitive learning and memory related areas,and can simultaneous trigger M1 receptor to increase central Ach contents. So,Huangjing Dilong extracts increase the activity of central cholinergic system in senile dementia's disease rats.
