摘 要:目的:观察芪附理中灌肠方对溃疡性结肠炎(UC)大鼠血清白介素-8(IL-8)、白介素-10(IL-10)及肠黏膜细胞间黏附分子-1(ICAM-1)表达的影响,以探讨其治疗UC的可能机制。方法:SPF级雄性大鼠36只随机分为模型组、空白组、柳氮磺吡啶组(0.32g/kg)及芪附理中灌肠方组(5.51,11.02,22.04g/kg)共6组。使用TNBS/无水乙醇建造UC大鼠模型,于造模后第8天开始给药,持续灌肠给药14天,观察芪附理中灌肠方灌肠对UC大鼠IL-8、IL-10及ICAM-1表达的影响。结果:与空白组相比。模型组大鼠肠黏膜损伤评分显著增高,较模型组而言,芪附理中灌肠方(22.04g/kg)量组肠黏膜炎症评分显著降低;与空白组相比,模型组大鼠血清IL-8、及肠黏膜ICAM-1含量显著增高,IL-10含量明显显著降低;与模型组相比,芪附理中灌肠方(22.04g/kg)量组血清IL-8及肠黏膜ICAM-1含量显著降低,IL-10含量明显增高。结论:芪附理中灌肠方对UC大鼠肠黏膜具有较好的抗炎修复作用,其机制可能与上调IL-10、下调IL-8及ICAM-1的表达有关。Objective: To observe the effect of Qifulizhong Enema on expressions of interleukin-8( IL-8),interleukin-10( IL-10) and Intercellular Adhesion Molecule-1( ICAM-1) of the rats with Ulcerative Colitis( UC). Methods: Thirty-six SPF male rats were randomly divided into the model group,the sulfasalazine group( 0. 32g/kg),the control group and the group of Qifulizhong Enema( 5. 51,11. 02,22. 04g/kg). The animal model was established by TNBS/ethanol,and started the medication 8 days after the establishment of the model. All rats were killed after clyster was kept for 14 days,the effect of Qifulizhong Enema was determined by detecting expressions of IL-8,IL-10 and ICAM-1 of UC rats. Results: Compared with the control group,the level of mucosal inflammation and damage score significantly increased in the model group. Compared with the model group,Qifulizhong Enema( 22. 04g/kg) significantly improved the level of mucosal inflammation and damage score,increased the expression of IL-10 and reduced expressions of IL-8 and ICAM-1( P〈0. 05). Conclusion: There is the anti-inflammatory repair effect of Qifulizhong Enema decoctions on UC rats' colonic mucosa. The possible mechanisms may be related to increasing the expression of IL-10 and reducing the expressions of IL-8 and ICAM-1.
