Effects of Modified Taohe Chengqi Decoction on Blood Glucose,Blood Lipid and Macrovascular Lesions in Diabetic Rats

摘  要:目的探讨加味桃核承气汤防治糖尿病大鼠大血管病变的可能机制。方法将80只雄性SD大鼠随机分为正常组、模型组、二甲双胍组、加味桃核承气汤1号方组(中药1组)、加味桃核承气汤2号方组(中药2组)。除正常组外,其余4组高糖高脂饮食6周后通过腹腔注射链尿佐菌素(STZ)40 mg·kg^(-1)建立糖尿病大血管病变模型。造模成功后,正常组和模型组每天采用等量蒸馏水灌胃,中药1组、2组分别采用加味桃核承气汤1号方、2号方5.4 g·kg^(-1)灌胃,二甲双胍组采用二甲双胍研磨溶液0.156 g·kg^(-1)灌胃。8周后处死大鼠,检测血糖、血脂变化情况,并采用免疫组化观察大鼠胸主动脉PI3K、Akt的染色情况,QRT-PCR法检测大鼠胸主动脉PI3K、Akt mRNA的表达。结果与模型组比较,中药1、2组均能显著降低血糖、血脂及PI3K mRNA、Akt mRNA水平(P<0.01),减少PI3K(p85)、Akt蛋白在大鼠胸主动脉的表达。其中,加味桃核承气汤2号方较1号方更能有效降脂、降糖(P<0.01);与二甲双胍组比较,中药2组能更有效降低PI3K mRNA、Akt mRNA水平,差异有统计学意义(P<0.01)。结论加味桃核承气汤能够有效防治糖尿病大鼠大血管病变,其机制可能与抑制PI3K/Akt信号通路,减少PI3K、Akt mRNA的表达以及降低血糖、血脂有关。此外,加味桃核承气汤2号方较1号方更能有效防治糖尿病大血管病变。Objective To explore the possible mechanism of modified Taohe Chengqi Decoction m preventxng cllabetlc macroangiopathy. Methods Eighty male Sprague-Dawley rats were randomly divided into normal group, model group, metformin group, modified Taohe Chengqi Decoction group 1 (TCM group 1), modified Taohe Chengqi Decoction group 2(TCM group 2). Ecept the normal group, the other 4 groups were fed by high-fat and high-carbohydrate diet for 6 weeks, then rats in these 4 groups were injected with streptozotoein(STZ, 40 mg·kg^-1) to establish diabetic macroangiopathy. After the model was successfully established, the rats in the normal group and the model group were treated with the same amount of distilled water every day. The TCM group 1 and the TCM group 2 were treated with modified Taohe Chengqi Decoction No.1 and modified Taohe Chengqi Decoction NO.2 (5.4 g·kg^-l·d^-1 both); metformin group were treated with metformin grinding slurry orally (0.156 g·kg^-l·d^-1). After 8 weeks, the rats were sacrificed and the blood glucose and blood lipid levels were detected. The expression of PI3K and Akt mRNA in the thoracic aorta of the rats were detected by qPCR. The expressions of PI3K and Akt protein in the thoracic aorta of rats were detected byimmunohistoehemistry. Results The levels of the blood glucose, blood lipids, PI3K mRNA and Akt mRNA in the thoracic aorta of the model group were significantly lower than those in the model group (P 〈 0.01 ), and the expression of PI3K(p85) and Akt protein in the thoracic aorta of the model group was significantly lower than those in the model group. Compared with the modified Taohe Chengqi Decoction NO.l, modified Taohe Chengqi Decoction NO.2 was more effective on decreasing blood lipid and glucose levels (P 〈 0.01 ); compared with the modified Taohe Chengqi Decoction NO.1 and metformin, modified Taohe Chengqi Decoction NO.2 also decreased the levels of PI3K mRNA and Akt mRNA(P 〈 0.01 ).
