摘 要:为了探析辣椒(Capsicum annuum)下胚轴不定根发生的分子机制,采用转录组学方法研究了不定根发生诱导期和启动期的差异表达基因。结果表明:在AR24/AR0、AR120/AR0和AR120/AR24比较组中分别鉴定到4 991、6 658和4 128个差异表达基因,其中1 321个为3个比较组所共有。对差异表达基因进行GO和KEGG显著性富集分析,发现辣椒下胚轴不定根发生的诱导期主要涉及激素应答、激素信号转导、细胞壁降解与重塑,启动期则是DNA复制、转录与翻译、细胞分裂和细胞壁合成等;纵观两个时期,不定根的发生由生长素、细胞分裂素、乙烯和油菜素内酯相关基因调控,并且与GRAS、AP2、ERF、LBD、SRS、WRKY家族转录因子表达密切相关。由上可知,辣椒下胚轴不定根发生是由一系列信号分子参与且极其复杂的器官发生过程。In order to investigate the molecular mechanism of adventitious rooting from pepper(Capsicum annuum) hypocotyl cuttings, we performed a genome-wide transcript analysis of rooting zone at 0, 24 and 120 h after planting. A total of 4 991, 6 658 and 4 128 differentially expressed genes(DEGs) were identified in AR24/AR0, AR120/AR0 and AR120/AR24 respectively, among which 1 321 DEGs were in common among the three comparison groups. The GO and KEGG enrichment analysis of DEGs were performed in this study. The results revealed that responses to hormones, hormone-mediated signaling pathways, and cell wall degradation and modification were involved in the induction phase of adventitious root development. While, DNA replication, transcription and translation, cell division, and cell wall biosynthesis were important events in the initiation phase. In addition, genes related to auxin, cytokinin, ethylene and brassinosteroid were differentially expressed during adventitious root development. Moreover, transcription factor gene families including GRAS, AP2, ERF, LBD, SRS and WRKY were probably important regulators of adventitious rooting from pepper hypocotyl cuttings. Taken together, adventitious rooting is a complicated organogenesis process regulated by diverse metabolic and signaling pathways.
