β-连环蛋白在艾灸肺俞、膏肓俞治疗肺纤维化大鼠中的表达变化

摘　　要：目的:观察β-连环蛋白(β-catenin)在艾灸肺俞、膏肓俞治疗肺纤维化大鼠中的表达的变化情况。方法:选用SD雄性大鼠32只,随机分为4组:正常对照组、模型组、艾灸治疗组、泼尼松治疗组。除正常对照组外,其余3组均采用博来霉素(5 mg/kg)灌入气管复制肺纤维化模型,治疗组从造模后第1天开始分别艾灸及泼尼松灌胃治疗,在造模后第11天取材。采用HE染色、Masson染色观察肺组织病理变化;分别采用免疫组化法和Q-PCR法检测β-catenin的表达水平。结果:艾灸组与模型组相比,肺组织病理程度明显降低,β-catenin表达显著降低,艾灸组与泼尼松组相比无明显差异。结论:艾灸治疗能降低β-catenin表达水平,降低肺泡炎程度及延缓肺纤维化进程。Objective： To observe the effect of the expression of β-catenin through moxibustion at Feishu（ BL13） and Gaohuangshu（ BL43） against the pulmonary fibrosis process of model rats. Methods： Thirty-two SD male rats were recruited and divided into four groups： normal control group,model group,moxibustion group and prednisone group. Pulmonary fibrosis model was made by the injection of bleomycin into the trachea,5 mg/kg. Since the second day after modeling,rats of the moxibustion group were given moxibustion at Feishu（ BL13） and Gaohuangshu（ BL43）. Rats of the prednisone group were given stomach-injection of prednisone every day. At 11 day after modeling,the rats were randomly selected and sacrificed. HE staining and Masson staining were used to observe the pathological changes of lung tissue. The expression of β-catenin was detected by immunohistochemistry and Q-PCR. Results： Compared with the model group,the degree of lung tissue pathology was significantly reduced in the moxibustion group and the prednisone group. There was no distint difference between the moxibusion group and the prednisone group.Conclusion： Moxibustion treatment can reduce the level of β-catenin,the degree of pulmonary inflammation and delay the progression of pulmonary fibrosis.
