摘  要:目的探讨黄芪多糖(APS)对大鼠蛛网膜下腔出血(SAH)后引起的早期脑损伤是否具有治疗作用。方法成年雄性SD大鼠120只,采用颈内动脉刺破法建立SAH模型,随机分为假手术(sham)组、SAH模型(SAH+NS)组、APS给药(SAH+APS)组。SAH模型建立后,各组立即腹腔注射相应溶液,24 h后通过T2加权成像(T2WI)来评估脑内损伤情况;应用Nissl染色法观察神经元特有结构尼氏体的形态学改变;应用免疫组织化学和Western blotting方法检测凋亡因子cleaved-Caspase-3和Bcl-2、Bax蛋白的表达水平。结果组织学染色及T2W1结果表明,SAH大鼠应用APS(40mg/kg)可以提高Bcl-2的表达水平(P<0.05),下调cleaved-Caspase-3和Bax的表达水平(P<0.05);T2WI结果显示,SAH大鼠应用APS(40mg/kg)可以降低脑内损伤程度(P<0.05)。结论黄芪多糖可以抑制SAH大鼠海马区神经元的凋亡,对早期脑损伤具有一定的治疗作用。Objective To investigate whether astragalus polysaccharide (APS) has a therapeutic effect on early brain injury induced by subarachnoid hemorrhage (SAH) in rats. Methods 120 adult male SD rats were randomly divided into sham operation (sham) group, SAH model group (SAH + NS) and APS group ( SAH + APS). After the establishment of the SAH model, each group was injected with the corresponding solution immediately. After 24hours, T2 weighted image (T2 weighted imaging, T2WI) was used to evaluate the brain injury;Nissl stainingmethod was applied to observe neuronal specific structure, morphological changes of austenite; Application of immunohistoehemistry and Western blottingmethod to detect apoptosis cleaved Caspase-3 and the Bel-2, Bax protein expression level. Results Histological staining and T2WI result showed that APS (40 mg/kg) could increase the expression of Bcl-2 (P 〈 0.05 ) , down-regulate the expression of cleaved Caspase-3 and Bax (P 〈0.05) The result showed that APS (40 mg / kg) could decrease the degree of brain injury ( P 〈 0.05 ). Conclusion Astragalus polysaccharide can inhibit the apoptosis of neurons in hippocampus of SAH rats, and has therapeutic effect on early brain injury.
