摘  要:目的:探讨胆固醇结石(GS)小鼠肝脏的核受体基因——肝核受体类似物1(LRH-1)及其调控基因BⅠ型清道夫受体(SRBI)的表达。方法:C57BL/6小鼠40只,均为雌性,10周龄,体重30~35 g。20只为一组,分别予以正常饮食(对照组)及高脂饮食(胆石组)喂养10周后处死。病理切片检查小鼠胆囊壁细胞组织学改变,实时定量PCR法测定肝脏LRH-I及SRBI的表达量。Western blot方法测定肝脏上述基因蛋白的表达量。结果:胆结石小鼠胆囊壁细胞呈现炎性改变。胆石组小鼠LRH-I mRNA及蛋白表达均高于对照组(P<0.01),SRBI mRNA及蛋白表达量较对照组升高(P<0.01)。结论:小鼠肝脏LRH-1基因的表达异常与胆囊胆固醇结石形成有关,同时SRBI的表达可能与结石形成有关,共同促进了小鼠胆结石的形成。Objective: To investigate the expressions of liver receptor homolog 1 (LRH-1) and scavenger receptor B type I (SRBI) gene in cholesterol gallstone (CGS) mice. Methods: Forty C57BL/6 mice, 20 with cholesterol gallstone (GS)and 20 controls without gallstones (GSF) were enrolled in this study, mRNA and protein expression of LRH-1 and SRBI genes were determined by RT-PCR and Western blot. Results: Gallbladder walls of GS mice were thicker with increased stromal granulocyte infiltration. The expression levels of LRH-1 genes were significantly higher in GS mice than in controls(P〈0.01). The expression levels of SRBI genes were also significantly higher in GS mice than in controls(P〈0.01). Conclusion: The increased expression of LRH-1 and SRBI gene may be related to GS disease.
