摘 要:鉴定鹅CYP2C45基因核心启动子序列,并筛选其单核苷酸多态性(single nucleotide polymorphism,SNP)位点,比较等位基因的转录活性差异及其与肝重性状的相关性,进而为肝重性状的选育筛选新的分子标记。通过双荧光素酶报告基因载体系统鉴定鹅CYP2C45基因启动子转录活性区域;通过PCR-SSCP、测序和序列比对方法分析朗德鹅的CYP2C45基因SNP,并分析不同基因型的转录活性;使用SPSS软件分析不同基因型与产肝性能的关系。结果表明通过双荧光素酶报告基因载体系统分析,发现鹅CYP2C45基因启动子活性区域为-165^+38bp;对朗德鹅群体进行了该区域SNP筛查,在5′上游-93bp处发现1个T/C突变,造成了屠体重的显著差异,对该突变造成的不同突变型进行转录活性分析,发现转录活性存在差异但未达到显著水平,推断CYP2C45基因的转录水平与鹅产肝性能呈正相关。本研究发现鹅CYP2C45基因核心启动子区域为-165^+38bp,该区域存在1个SNP,且与屠体重显著相关。The objective of this study is to identify the core promoter sequence of the goose CYP2C45(cytochrome P450 2C45)and screen single nucleotide polymorphism(SNP)loci,analyze and compare the different transcriptional acticity of the allele and their correlation with the liver weight.And then select the new molecular markers for breeding of liver weight liver weight.The transcription activity of CYP2C45 gene promoter was detected by dual luciferase reporter gene vector system.The SNPs were analysed by PCR-SSCP,sequencing and alignment.The transcription activities of different genotypes were analyzed by dual luciferase reporter gene vector system.The correlation of genotype and goose fatty liver performance was analyzed by SPSS.The results show that through dual luciferase reporter gene vector system,the active region of CYP2C45 gene is found at-165^+38bp of5'flanking.One SNP,a T/C mutation at-93 bp upstream of 5'flanking,is obtained and resulting significant difference of dressed weight.The transcription activities of different mutation types are different but not reach a significant level.It is deduced that there is a positive correlation between transcription activity and goose fatty liver performance.The core promoter region of CYP2C45 gene is-165^+38bp.There is a significant difference between dressed weight and the SNP found in this region.
