摘 要:目的采用响应曲面法的Box-Behnken效应面法建立数学模型,优化夏枯草中熊果酸的超声提取工艺。方法以料液比、超声提取时间、乙醇体积分数为考察因素,采用反向高效液相色谱法检测不同条件下夏枯草中熊果酸的含量。结果夏枯草中熊果酸的最优提取工艺为:料液比(g·mL^(-1))1∶5,超声提取时间1.5 h,乙醇体积分数100%,此工艺条件下,熊果酸的含量为0.623 2 mg·g^(-1),与预测值0.611 8 mg·g^(-1)相差较小。结论通过响应曲面法得到了夏枯草中熊果酸提取的优化条件,响应曲面法准确、可靠、可行性强、实用性好。Objective To establish mathematical model by Box-Behnken response surface method of response surface method,and optimize the ultrasonic extraction process of ursolic acid in Prunella vulgaris L.Methods The content of ursolic acid in Prunella vulgaris L.was detected by reverse-high performance liquid chromatography( HPLC) with the ratio of material to liquid,ultrasonic extraction time and ethanol volume fraction.Results The optimum extraction process for ursolic acid from Prunella vulgaris L.: solid-liquid ratio( g·mL^-1) was 1 ∶ 5,ultrasonic extraction time was 1.5 h,ethanol concentration was 100%.Under this process,the content of ursolic acid was 0.623 2 mg·g^-1,which was less than the predicted value of 0.611 8 mg·g^-1.Conclusion The optimized conditions of extraction of ursolic acid in Prunella vulgaris L.were obtained by response surface methodology.The response surface method was accurate,reliable,feasible and practicable.
