药物制剂中月桂醇磷酸酯钾的新型处理方法及对水溶性药物成分表征的影响

摘  要:目的:建立一种新型、简便的方法处理药物制剂中的月桂醇磷酸酯钾(HR-S1),并对其中的水溶性药物成分进行含量表征。方法:取润肤霜2.0g,加入甲醇10 mL,搅拌,过滤,加甲醇5 mL洗涤残渣,合并滤液;加入0.25 mL磷酸,摇匀,用10 mL正己烷萃取,重复3次,去除正己烷层,残余溶液氮吹至干,加甲醇复溶。润肤霜中人参皂苷Rg1的检测:C18色谱柱为固定相,乙腈-0.1%醋酸梯度洗脱,检测波长为203 nm。结果 :本研究所建立的方法能完全去除HR-S1对润肤霜中水溶性药物成分人参皂苷含量表征的干扰,方法回收率高至99.6%,精密度和重复性好。结论:该方法操作简便快速,能完全消除HR-S1的干扰,回收率高,可用于乳霜中月桂醇磷酸酯钾的去除及人参皂苷Rg1的表征。OBJECTIVE To establish a new method to deal with the lauryl phosphate potassium in pharmaceutical preparations and characterization of the water soluble ingredient in pharmaceutical preparations.METHODS Precisely 2.0g sample was dissolved in 10 mL of methanol,then the solution was filtered,5 mL of methanol was added to dissolve residue,0.25 mL of phosphoric acid was added into the incorporated filtrate to shake and extract with 10 mL n-hexane.The procedure was repeated for three times,lauryl phosphate potassium was removed,and the residual solution was dried by nitrogen and reconstituted by methanol.Ginsenosides Rg1 detection:using C18 chromatographic column as stationary phase,acetonitrile and 0.1%acetic acid for gradient elution at a detection wavelength of 203 nm.RESULTS The method completely eliminated the interference of lauryl phosphate potassium on water soluble ingredient namely ginsenosides Rg1 in creams.The recovery rate of the method was up to 99.6%,with good precision and repeatability.CONCLUSIONthe method is simple and rapid,has a high recovery,can completely eliminate the interference of HR-S1,and can be used to remove lauryl phosphate potassium and characterize ginsenosides Rg1.
