摘 要:为选育抗疱疹病毒(Cy HV-3)的新品种德国镜鲤,本研究在F_1抗病家系基础上构建10个家系分A、B两组混养。从193对引物中分别筛选3对高多态性微卫星引物进行亲子鉴定及遗传结构分析。结果显示:(1)两组中3个位点的累积排除率为99.99%、99.98%,亲子代配对率为100%(p=0.05),说明利用3个高多态性位点就可达到亲子鉴定的目的。(2)家系鉴定成活率为0%~60.21%。其中1#、8#、9#、10#家系比对照组提高43.51%、22.00%、11.64%、8.45%,可用于F_3选育。(3)1#、8#、9#、10#4个家系的遗传多样性指标为:Ae为3.507 7~4.033 2、Ho为0.818 1~0.965 8、He为0.7062~0.7386,遗传多样性水平较高。(4)除9#家系的HLJ3939位点显著偏离Hardy-Weinberg平衡(p<0.05)外,其他位点均极显著偏离Hardy-Weinberg平衡(p<0.01),说明所选位点均经过了选择作用。(5)1#、8#、9#、10#4个家系FST为0.057 3~0.115 6,8#、9#、10#3个家系间的Nei氏遗传距离均值为0.515 0,说明4个家系具有较高遗传潜力。因此,本研究成功筛选出4个成活率较高且具有较高遗传潜力的家系,以用于下一代选育。In order to breed new species of German mirror carp which was resistant to Cy HV-3,we constructed ten families and cultured them mixedly into two groups(A and B) based on the F_1 disease-resistant family.We screened out three pairs of high polymorphic microsatellite primers from 193 pairs of primers for paternity identification and genetic structure analysis.The results show that:(1) Cumulative elimination rate of 3 loci in the two groups were 99.99% and 99.98%,respectively.Parents-offspring pair rate was 100%(p=0.05),which indicated that three high polymorphisms loci could reach the requirements of paternity identification.(2) The survival rates of each family were 0% ~60.21%.The survival rate of family 1#,8#,9# and 10# increased by43.51%,22%,11.64% and 8.45% compared to the control group,and they could be used for F_3 breeding.(3)The genetic diversity indexes of 1#,8#,9# and 10# these four families:Ae ranged from 3.507 7 to 4.033 2,Ho from0.818 1 to 0.965 8,and He from 0.706 2 to 0.738 6.The genetic diversity levels of those families were relatively high.(4) Only the HLJ3939 locus of 9# family was significantly deviated from Hardy-Weinberg equilibrium(p〈0.05).Other loci were extremely significantly deviated from Hardy-Weinberg equilibrium(p〈0.01).This meant that all the loci had experienced selection.(5) The FST of 1#,8#,9# and 10# family ranged from 0.057 3 to 0.115 6,and the mean value of Nei's genetic distance among family 8#,9# and 10# was 0.515 0.It showed that those four families had relatively high genetic breeding potential.Therefore,we successfully screened out 4 families with relatively high survival rate and genetic breeding potential for the next generation of breeding.
