循经取穴针刺对心肌缺血大鼠电压门控氯通道相关基因表达的影响

摘　　要：目的:对比观察电针对心肌缺血大鼠氯离子通道相关基因表达的影响,探讨循经取穴针刺治疗心肌缺血的作用机制。方法:70只雄性SD大鼠随机分为空白组10只,模型组、内关穴组、列缺穴组及非经非穴组各15只。采用皮下异丙肾上腺素注射复制心肌缺血模型。电针治疗组每日治疗1次,每次20 min。针刺干预7 d后,采用Western Blot法和Real-time PCR法检测蛋白激酶C(PKC)、蛋白激酶G(PKG)以及电压门控氯离子通道相关基因(Cl C-2、Cl C-3)的表达。结果:与空白组比较,模型组大鼠心肌PKC、PKG、Cl C-2、Cl C-3升高(P<0.05);与模型组比较,各组大鼠心肌组织PKC、PKG、Cl C-2、Cl C-3表达量均显著降低(P<0.05);与内关组比较,列缺组与非经非穴组上述基因表达水平的差异有统计学意义(P<0.05)。结论:各组PKC、PKG、Cl C-2、Cl C-3的表达差异反映了心肌缺血氯通道的响应特性,提示内关穴对心肌缺血保护作用的循经特异性效应。Objective：To compare the effects of needling acupoints along meridians on gene exprssion of chloride channel in rats with myocardial ischemia so as to explore its protection mechanism of ischemic myocardium. Methods ： Seventy SD rats were randomly divided into blank control group, model control group, Neiguan〈 PC6）, Lieque （LU7）and non- acupoint groups. All rats were injected isoproterenol （ISO） to establish myocardial ischemia rats model except for the blank control group. All the treatment groups were received electro - acupuncture therapy for 20 min daily and lasted for a week. And then, detect the expression levels of PKC and PKG by Western Blot method and the chloride channel＇ s gene by real - time quantitative PCR. Results ： In comparison with the model control group, the expression levels of PKC, PKG, ClC -2 and ClC -3 in all the treatment groups were significantly down -regulated（ P 〈 0.05 ）. In comparison with the other treatment groups, there was statistical significance in the expression levels of the above experiment indicators in the Neiguan （ PC6 ） group. Conclusion ： The results showed some characters of chloride channel response, indicating that Neiguan acupoint might be tightly related to the heart on myocardial ischemia rats along the Hand -Jueyin pericardium meridian.
