三种针法对对氯苯丙氨酸诱导失眠大鼠海马5-HT_(1A)、5-HT_(2A)基因表达影响研究  被引量:18

摘  要:目的:观察针刺失眠方、脐内环穴,失眠方和脐内环穴联合应用对对氯苯丙氨酸(para-chlorophenylalanine,PCPA)失眠大鼠海马5-HT_(1A)、5-HT_(2A)总RNA表达的影响,并对三种针法的效果进行比较研究。方法:随机抽取6只正常大鼠作为正常组,将30只PCPA失眠大鼠随机分为模型组、针刺失眠方组、脐内环针组、失眠方加脐内环针组、非穴组5组,每组6只。正常组、模型组无特殊处理,干预组分别给予针刺失眠方、针刺脐内环穴、针刺失眠方和脐内环穴联合干预,非穴组予针刺非穴处理,治疗6 d后取大鼠海马组织,采用real-time PCR方法检测5-HT_(1A)、5-HT_(2A)的基因表达。结果:PCPA失眠大鼠海马5-HT_(1A)基因表达下降,而5-HT_(2A)基因表达上调;三种针法均显著上调PCPA失眠大鼠海马5-HT_(1A)的基因表达、下调5-HT_(2A)的基因表达;而失眠方和脐内环穴联合应用的效果优于失眠方或者脐内环穴单用。结论:PCPA失眠大鼠可能存在中枢5-HT受体信号机制异常,三种针法可能通过调节海马内5-HT_(1A)、5-HT_(2A)基因表达而改善失眠;实验支持失眠方和脐内环穴针刺联合应用为失眠症的优选干预方法。Objective: To observe the three acupunture methods effects(the acupuncture of umbilical inner acupoints, the acupoint-prescription of insomnia, their combined method) on the total RNA expressions of 5 -HT1A and 5 -HT2A in hippocampus of PCPA insomnia model rats and select the optimal acupuncture method. Methods : Thirty PCPA insomnia male rats were randomly divided into five groups, and there was still another normal group. Each of the six groups had 6 rats. The normal and the model groups were given no special treatment. The observation groups were given acupuncture treatment of the umbilical inner acupoints, the acupoint - prescription of insomnia, their combined method and the insomnia acupoint - prescription respectively. The non - acupoint group was dealt with non - acupuncture. After six days of treatment, in the hippocampus of the rats, the gene expression levels of 5 - HT1A and 5 - HT2A were detected by real - time PCR. Results: The gene expression level of 5 -HT1A in hippocampus of PCPA insomnia model rats was significantly reduced, and that of 5 - HT2A was significantly elevated. Three acupuncture methods can improve the gene expression levels of 5 - HT1A and reduce the gene expression levels of 5 - HT2A in hippocampus of rats. Their combined method and the insomnia acupoint - prescription were better than each single method of acupuncture. Conclusion : The PCPA insomnia rats exist the abnormal information mechanism of central 5 - HT receptor. Three acupuncture methods might take effects on insomnia by the mechanism of improving the gene expression level of 5 - HT1A and reducing the gene expression levels of 5 - HT2A in hippocampus. The experiment suggested that the combined method of the acupoint - prescription of umbilical inner acupoints and the insomnia acupoint- prescription therefore be the preferred acupuncture.
