摘 要:目的:比较吴茱萸不同炮制品体外对人正常肝细胞L02的毒性作用,初步探讨吴茱萸炮制减毒的原理。方法:以人正常肝细胞L02细胞为研究对象,采用CCK-8法检测吴茱萸不同炮制品对L02细胞活性的影响,采用倒置显微镜观察细胞形态学改变,并测定细胞培养上清液中谷丙转氨酶(ALT),谷草转氨酶(AST)和乳酸脱氢酶(LDH)的含量。结果:与阴性对照组比较,阳性药组、生吴茱萸组可明显降低L02细胞的活性(P<0.01)与形态,显著升高L02细胞培养上清液中ALT、AST、LDH活力(P<0.01,P<0.05);与生吴茱萸组比较,甘草制吴茱萸组、盐吴茱萸组可显著降低对L02细胞的增殖抑制作用(P<0.01)与形态变差的趋势,显著降低L02细胞培养上清液中ALT、AST、LDH活力(P<0.01,P<0.05)。吴茱萸各炮制品体外对L02细胞的毒性作用顺序为:生吴茱萸>甘草制吴茱萸>盐吴茱萸。结论:生吴茱萸、甘草制吴茱萸、盐吴茱萸体外对L02细胞具有毒性作用,炮制可降低吴茱萸的体外肝毒性作用。Objective : To compare the virulence of different processed products of Evodia rutaecarpa ( Juss. ) Benth on L02 human normal liver cellsin vitro and study the attenuation of Evodia rutaecarpa ( Juss. ) Benth in different processed ways. Method: Take L02 human normal liver cells as research object and test the effect to activities of L02 cells with the method of CCK - 8 by processing Evodia rutaecarpa ( Juss. ) Benth in different ways. Observe the change of cell morphology with inverted microscope and determine the content of alanine aminotransferase (ALT) , aspartate aminotransferase (AST) and lactate dehydrogenase (LDH) in culture supernate. Result: Compared with the negative group, the positive group and raw product group of Evodia rutaecarpa ( Juss. ) Benth can obviously reduce the activities of L02 cells ( P 〈 0. 01 ) and morphology. Additionally, the activities of ALT, AST and LDH were promoted markedly ( P 〈 0. 01, P 〈 0. 05). Compared with the raw product group of Evodia rutaecarpa (Juss.) Benth, glycyrrhiza processed group and salt processed group can observably inhibit the proliferation of L02 cells(P 〈0. 01 )and the trend on morphological deterioration. Moreover, the activities of ALT, AST and LDH were obviously decreased(P 〈 0. 01 ,P 〈 0. 05). The toxic effects of different processed products of Evodia rutaecarpa (Juss.) Benth on L02 cells in vitro was the raw product 〉 the glycyrrhiza processed 〉 the salt processed. Conclusion : The raw products,glycyrrhiza processed products and salt processed products of Evodia rutaecarpa ( Juss. ) Benth were toxic of L02 cells in vitro and hepatotoxicity could be reduced by processed.
