摘  要:牛白血病是由牛白血病病毒(Bovine leukemia virus,BLV)引起的牛(Bos taurus)、绵羊(Ovis aries)等动物的慢性肿瘤性疾病,是影响养牛业发展的重要传染病之一。为探索牛趋化因子受体1(chemokine(CX-C motif)receptor 1,CXCR1)基因编码区(coding sequence,CDS)SNPs与牛白血病易感性之间的关系,本研究用荧光共振能量转移-定量多聚酶链式反应(fluorescence resonance energy transfer-quantitative polymerase chain reaction,FRET-qPCR)对某大型奶牛场866头中国荷斯坦牛牛白血病感染情况进行检测,同时用飞行时间质谱法(matrix-assisted laser desorption/ionization time of flight mass spectrometry,MALDI-TOF-MS)对CXCR1基因编码区CXCR1-c.642 A>G、CXCR1-c.816C>A、CXCR1-c.980A>G和CXCR1-c.1068G>A 4个SNPs位点进行检测,采用Logistic回归分析CXCR1 SNPs基因型与牛白血病易感性之间的关系,结果表明:在866头奶牛中,患白血病的牛为42头,阳性率为4.85%;CXCR1-c.642 A>G、CXCR1-c.816C>A、CXCR1-c.980A>G和CXCR1-c.1068G>A 4个SNPs位点优势基因型分别为GG、CC、AA和GG型,频率分别为0.580、0.466、0.60和0.658。CXCR1-c.980 A>G位点对牛患白血病的相对风险影响达到显著水平(P=0.016)。CXCR1-c.980 A>G位点GG型基因患白血病的概率为AA型基因的5.04倍。而CXCR1-c.642 A>G、CXCR1-c.816C>A、CXCR1-c.1068G>A和CXCR1单倍型对牛白血病易感性无显著影响(P>0.05)。CXCR1c.980A>G位点可作为奶牛白血病抗病育种的分子标记之一,筛选CXCR1-c.980A>G位点AA型个体有助于降低奶牛对牛白血病的易感性并提高生产效率。Bovine leukemia is a kind of chronic tumor disease caused by Bovine leukemia virus (BLV) in cattle (Bos taurus), sheep (Ovis aries) and other animals. In order to verify whether there is an association between BLV and SNPs of CXCR1 gene coding sequence (CDS), bovine leukemia infection of 866 Chinese Holstein was detected by fluorescence resonance energy transfer- quantitative polymerase chain reaction (FRET- QPCR) in this study. The SNPs (CXCR1-c.642A〉G, CXCR1-c.816C〉A, CXCR1-c.980A〉G and CXCR1- c. 1068G〉A) in the CDS of CXCR1 gene were detected by MALDI-TOF-MS (matrix-assisted laser desorption/ionization time of flight mass spectrometry). The relationship between genotype and haplotype of CXCR1 SNPs and bovine leukemia infection was analyzed by Logistic regression. The results showed: 1) Of the 866 cows, 42 had bovine leukemia, with a positive rate of 4.85%. 2) The predominant genotypes of CXCRl-c.642 A〉G, CXCR1-c.816C〉A, CXCR1-c.980A〉G and CXCR1-c.1068G〉A were GG, CC, AA and GG, with frequencies of 0.580, 0.466, 0.60 and 0.658, respectively. The relative risk of leukemia was significantly affected by CXCRl-c.980A〉G locus (P=0.016). The individual of GG genotype was 5.04 times suffering from leukemia than that of AA genotype. CXCRl-c.642A〉G, CXCRI-c.816C〉A, and CXCRI-c.lO68G〉A have no significant effects on the susceptibility to bovine leukemia. CXCR1-980 A〉G can be used as a marker for disease resistance breeding of dairy cows, and selecting AA genotype individuals can help to reduce the susceptibility of bovine leukemia and improve productivity efficiency.
