有瓜石斛水煎液HPLC特征图谱研究及其与药材的相关性分析

摘  要:目的建立有瓜石斛水煎液HPLC特征图谱,并与药材进行相关性分析。方法采用ZORBAX SB C18色谱柱;流动相为乙腈-0.1%磷酸溶液(梯度洗脱);检测波长分别为220nm、340nm;柱温为35℃;流速为1.0m L·min-1。结果在220nm检测波长下,11批有瓜石斛共标示出21~25个特征峰,可分为3类,均检出17个特征共同峰。其中第Ⅰ类5批有21个共有峰,相似度为0.906~0.972;第Ⅱ类3批有25个峰,相似度为0.955~0.986;第Ⅲ类3批有21个峰,相似度为0.979~0.989;同类样品的相似度为0.906~0.989;不同类样品共有模式间的相似度仅为0.481~0.763,有明显差异。在340 nm检测波长下,11批有瓜石斛共标示出7个黄酮类特征峰,相似度达0.909~0.985。两种检测波长条件下,有瓜石斛煎液基本能检出原药材相应特征峰,但相对峰面积比值有一定差异;每批药材与其煎液之间的相似度为0.733~0.919。结论方法准确可靠,重复性好,提示常规水煎煮工艺能保留有瓜石斛的主要特征成分,为评价有瓜石斛水煎液物质基础提供参考依据。OBJECTIVE To establish the characteristic spectrum for Flickingeria fimbriata decoction by HPLC and analyze the correlation of the decoction pieces and decoction of Herba flickingeriae. METHODS HPLC was performed on ZORBAX SB C18 column,the acetonitrile-phosphoric acid solution (φ =0. 1% ) was employed as a mo- bile phase by gradient elution,detection wavelength was 220nm and 340nm;column temperature was 35℃ ,and flow rate was 1.0mL ·min-1. RESULTS The 11 batches of Flickingeria fimbriata decoction were divided into 3 groups at the detection wavelength of 220nm,21 ~ 25 specific peaks were separated and 17 of them were the common specif- ic peaks for the 3 groups. The similarities were 0. 906 - 0. 972 with 21 common peaks in the 5 batches of first group, 0. 955 ~0. 986 with 25 common peaks in the 3 batches of second group, and 0. 979 ~ 0. 989 with 21 common peaks in the 3 batches of third group. The similarities were 0. 906 ~ 0. 989 in the samples of the same groups, while were only 0. 481 -0. 763 in the samples between the groups. At the detection wavelength of 340nm,the similarities of 11 batches were 0. 909 -0. 985 with 7 common peaks of flavonoids. The corresponding characteristic peaks were mostly detected in the decoction at the detection wavelength of 270nm and 340nm, though the peak area ratio of each com- mon peak had some differences. The similarity between the decoction pieces and decoction was 0. 733 ~ 0. 919. CONCLUSION The method is accurate and reliable,and has good reproducibility. The main characteristic components of Flickingeria fimbriata could be extracted by traditional decoction method, which will provide a certain reference to the material base of Flickingeria fimbriata decoction.
