摘 要:目的:观察交泰丸对2型糖尿病(T2DM)db/db小鼠肾脏的保护作用以及对脂肪组织中PPARγ基因表达的影响。方法:db/db小鼠分为模型组、交泰丸组和罗格列酮组,另取db/m小鼠为正常对照组;每组8只。给药处理4周,观察小鼠的饮食摄入量、尿量、体质量、空腹血糖(FBG)、血清三酰甘油(TG)、总胆固醇(TC)以及尿液指标变化,取肾脏进行HE染色观察其病理改变,RT-PCR检测脂肪组织PPARγmRNA表达。结果:交泰丸能够显著改善db/db小鼠饮食摄入量、尿量、FBG、血清三酰甘油(TG)以及尿液指标(P<0.05);上调脂肪组织中PPARγ的基因表达水平(P<0.05);减轻db/db小鼠的肾组织损伤。结论:交泰丸可有效地改善db/db小鼠的糖尿病症状,而且可以在不增加体质量的同时上调PPARγ的基因表达,修复db/db小鼠的肾损伤。Objective: To explore the protective effect of Jiaotai Pill on type 2 diabetes mellitus (T2DM) of db/db mice kidney and the regulation effect on PPAR gene expression in of adipose tissue. Methods: db/db mice were divided into model group, Jiaotai Pill group and rosiglitazone group, and db/m mice were set as nor- mal control group, each contain 8 mice. Each group was given the corresponding medicine or saline for 4 weeks, food and water intake, urine volume, body weight, fasting blood glucose (FBG) , blood lipid and urine indexes were detected, the kidneys were stained with HE to observe the pathological changes and the expression of PPARγ/mRNA in adipose tissue was detected by RT- PCR. Results: Jiaotai Pill significantly improved the food intake, water intake, urine volume, FBG, triglyceride (TG) and urine indexes of db/db mice (P 〈0.05) ; up -regulated the expression on PPARγ/in adipose tissue( P 〈 0.05 ) ; protected renal tissue from injury. Conclu- sion: Jiaotai Pill not only can effectively improve the symptoms of diabetes in db/db mice, but also can inhibit the weight gain and increase PPARγ/mRNA expression, repair the kidney damage in db/db mice.
