通络散结凝胶外用对骨癌痛大鼠痛觉行为及骨密度的影响

摘　　要：目的探讨通络散结凝胶外用对骨癌痛的影响。方法将30只大鼠随机分为假手术组、中药组和安慰剂组,每组10只。中药组和安慰剂组采用注射肿瘤细胞方法建立大鼠骨癌痛模型。假手术组和安慰剂组大鼠造模侧胫骨局部外用空白凝胶,治疗组外用通络散结凝胶。各组均于每日8:00、20:00给药1次,每次1 g,各组均给药21天。观察大鼠的一般状况、体重、自发性疼痛评分、热痛觉、机械痛觉、患肢承重、胫骨骨密度变化。结果与假手术组比较,安慰剂组和中药组活动逐渐减少,被毛光泽度降低,体质量下降。中药组大鼠体重、热痛觉缩足潜伏期、机械性痛觉50%缩足阈值、患肢承重、胫骨骨密度在第16~21天时显著高于安慰剂组(P<0.05或P<0.01),中药组造模第16~20天自发性疼痛评分低于安慰剂组(P<0.01),但造模第10、16、20天自发性疼痛评分高于假手术组(P<0.01)。结论通络散结凝胶外用能显著改善骨癌痛大鼠一般状况、体重及自发性疼痛、机械痛和热痛觉、患肢承重及骨密度。Objective To investigate the effect of externally used Tongluo Sanjie Gel （通络散结凝胶） on bone cancer pain.Methods Thirty rats were randomized into sham operation group,Chinese medicine group and placebo group,with 10 mice in each group.Tumor cells were injected to establish rat bone cancer pain model in Chinese medicine group and placebo group.The rats in sham operation group and placebo group were externally administered blank gel on the model side of the tibia,while Tongluo Sanjie Gel was used in the treatment group.Each group was given the gel twice a day respectively on 8： 00 and 20： 00,with 1g gel each time.The treatment lasted for 21 days.The general condition,weight,spontaneous pain score,thermal pain,mechanical pain,injured limb bearing and tibial bone mineral density （BMD） were observed.Results Compared with those of the sham group,the activity of the placebo group and the Chinese medicine group decreased gradually,the hairiness decreased and the body weight decreased.The bone weight,paw withdrawal thermal latency （PWTL） and 50% paw withdrawal mechanical threshold （PWMT）,injured limb bearing and tibial BMD in the Chinese medicine group was significantly higher than those in the placebo group from day 16 to day 21 （P 〈 0.05 or P 〈 0.01）.The spontaneous pain score in the Chinese medicine group was lower than that in the placebo group from day 16 to day 20 （P 〈 0.01）,but the scores on day 10,day 16 and day 20 were higher than those in the sham operation group （P 〈 0.01）.Conclusion Externally used Tongluo Sanjie Gel can significantly improve the general condition,weight,spontaneous pain,mechanical pain and thermal pain,injured limb bearing and BMD of rats with bone cancer pain.
