摘  要:目的:从细胞水平初步探讨TLR_4及其配体对矽肺晚期炎症的作用。方法:(1)制备小鼠矽肺模型:染尘采用滴鼻注入SiO_2悬液方法,剂量为200mg/kg,每只注入量为0.1mL。对照组在相同条件下滴鼻注入生理盐水0.1mL。(2)指标检测:Western blot检测成纤维细胞内TLR_4和RAGE蛋白含量。结果:(1)TLR_4结果显示:TLR_4表达水平随观察时间的延长明显升高;(2)RAGE结果显示:RAGE在正常对照组呈现高表达,在3d染尘时段表达量最高且均高于正常对照组,其余染尘组表达量降低。结论:在矽肺晚期炎症的形成过程中,HMGB1发挥生物学效应不完全依赖RAGE受体,还有TLR_4的参与。Objective:To explore the effects of TLR_4 and its ligands on late silicosis from the cell level.Methods:(1)Silicosis model preparation:SiO_2 suspension was injected with the method of nasal drip,with a dose of 200 mg/kg,each injection volume of 0.1mL.The control group was injected saline solution of 0.1mL under the same condition.(2)Index detection:The content of TLR_4 and RAGE were detect by Western blot.Results:(1)TLR_4 results showed that the expression level of TLR_4 was significantly increased with the prolongation of observation time(P0.05).(2)The results of RAGE showed that RAGE presented high expression in the normal control group;and the expression level was the highest in the three days of dusting day while the expression of the remaining dusting groups decreased(P0.05).Conclusion:In the formation of late silicosis inflammation,the biological effects of HMGB1 do not rely solely on the RAGE receptor,but also on the participation of TLR_4.
