摘  要:目的 探究胰岛素样生长因子1(IGF-1)转染自体骨髓间充质干细胞移植在兔胫骨牵张成骨模型实验研究中是否具有促进牵张成骨骨形成作用.方法 健康新西兰白兔48只,随机分为三组,每组16只.建立兔胫骨牵张成骨模型,实验组于固定期第2天于牵张成骨间隙内植入IGF-1基因介导转染的自体骨髓间充质干细胞.对照组同时间在牵张成骨间隙内植入相同浓度自体骨髓间充质干细胞.空白组牵张间隙内不注射任何物质.结果 在固定期2周和6周,实验组牵张区在X线摄片结果、HE染色观察方面,成骨质量明显优于对照组和空白组.3个月后取各组牵张成骨区标本做骨密度测定及生物力学测试,结果显示实验组以上两指标均明显优于对照组和空白组.结论 IGF-1转染的自体骨髓间充质干细胞移植能有效提高兔胫骨牵张成骨骨形成质量.Objective To study the effect of bone marrow mesenchymal stem cells (MSCs) transplantation insulin like growth factor 1 (1GF-1) on the distraction osteogenesis in rabbit tibia model in the experimental study. Methods Prepare healthy New Zealand white rabbit 48, and were randomly divided into three groups, each group have 16. Established rabbit tibia distraction osteogenesis model, the experimental group was treated by autologous bone marrow mesenchymal stem cells transplantation of transfected by insulin like growth factor 1 gene. On the 2nd day, the same concentration of autologous bone marrow mesenchymal stem cells were implanted in the control group at the same time. The blank group was not injected with any substance in the gap. Results In a fixed period the 2th and 6th week, in the X-ray beam results, HE staining observation aspect osteogenesis quality were better in experimental group than control group and blank group. 3 months later bone mineral density and biomechanical tests were performed in each group these two indexes were obviously better in experimental group than other two groups. Conclusion 1GF -1 transfected autologous bone marrow mesenchymal stem cells transplantation can effectively improve the quality of bone formation in rabbit tibia distraction osteogenesis.
