摘 要:目的 通过体外形态学观察、生物力学及免疫原性检测实验,探究不同化学萃取次数对同种异体肌腱生物活性的影响.方法 选取体重2.5~3.0 kg的雄性新西兰大白兔16只,随机分为四组,每组4只,A组(萃取一遍组)、B组(萃取二遍组)、C组(萃取三遍组)及D组(空白对照组).将其跟腱取材后分别进行相应次数的化学萃取过程,然后再分别进行HE染色、电镜观察、生物力学检测及混合淋巴细胞培养测定.结果 A,B两组肌腱表面色泽及纤维束与空白对照组无明显差别,C组与空白对照组差别较大;四组间生物力学检测结果无明显差异;四组间淋巴细胞转化率结果差异有统计学意义.结论萃取二遍组肌腱在去除免疫原性的同时,较完整地保留了大部分原有的肌腱纤维,具有良好的生物学性能.Objective To explore the the biological influence of different times of chemical extraction on the allogeneic tendon by exoteric morphological observation, biomechanics and detection of immunogenicity. Methods Sixteen New Zealand rabbits (weight 2.5~3.0 kg) were randomly divided into four groups, group A (chemical extraction once group), group B (chemical extraction twice group), group C(chemical extraction three times ) and group D ( blank control group), with 4 rabbits in each group. The Achilles tendon of each rabbit was separated and undertaken chemical extraction process, then HE dyeing, electron microscopy observation, biomechanical testing and mixed lymphocyte culture measured were conducted. Results There were no significant differences in the surface color and fiber bundle between A, B group and blank control group, but group C had bigger differences with blank control group. The four groups had no significant differences in the biomechanical testing. The four groups had significant differences in the lymphocyte culture measured. Conclusion Extraction twice group tendon retain the most original tendon fiber ntegrallty when removing immunogenicity, and also has good biological properties.
