摘　　要：目的探讨集落刺激因子1受体(CSF-1R)对人鼻咽癌6-10B细胞细胞周期素(cyclin)D1表达及其增殖的影响。方法选取人鼻咽癌6-10B细胞,体外利用慢病毒构建的CSF-1R过表达载体LV-CSF1R(16957-1)转染到人鼻咽癌6-10B细胞中(转染组),未转染CSF-1R慢病毒载体的鼻咽癌6-10B细胞为对照组。采用RT-q PCR法及Western blotting法检测转染组及对照组细胞CSF-1R、cyclin D1的表达情况,CCK-8法检测各组细胞活力的改变情况。结果转染组细胞CSF-1R mRNA及其蛋白表达水平明显高于对照组(P均<0.05)。转染组鼻咽癌6-10B细胞活力较对照组明显增高(P<0.05)。结论过表达CSF-1R可以通过调节细胞周期蛋白cyclin D1来促进鼻咽癌6-10B细胞的恶性生长。Objective To investigate the effects of colony stimulating factor-1 receptor（ CSF-1R） on cyclin D1 expression and cell proliferation of human nasopharyngeal carcinoma 6-10 B cells. Methods The human nasopharyngeal carcinoma 6-10 B cells were transfected with CSF-1R lentiviral vector LV-CSF1R（ 16957-1）（ transfection group）,and the 6-10 B cells without transfection of CSF-1R lentiviral vector were taken as the control group. The expression of CSF-1R and cyclin D1 was detected by RT-q PCR PCR and Western blotting. CCK-8 assay were used to detect the cell activity in each group. Results The expression of CSF-1R at mRNA and protein levels increased significantly in the transfection group as compared with that of the control group（ P〈0. 05）. The activity of 6-10 B cells was significantly higher in the transfection group than that of the control group（ P〈0. 05）. Conclusion Over-expression of CSF-1R significantly promotes the malignant growth of nasopharyngeal carcinoma 6-10 B cells by regulating the expression of cyclin D1.
