摘  要:目的将逆转录聚合酶链式反应(RT-PCR)应用于沙眼衣原体药物敏感性实验,对沙眼衣原体的药敏实验进行优化。方法将经Mc Coy细胞培养法检测出的40例沙眼衣原体临床株,传代培养至感染率达90%以上,进行多西环素的药敏实验。采用传统碘染方法和RT-PCR法来测定多西环素药物的MIC值。采用t检验比较两种方法的区别。结果采用RT-PCR测定的MIC值要高于传统碘染方法(P<0.05)。结论用RT-PCR代替传统碘染方法能够提高沙眼衣原体体外药物敏感实验的敏感性,有助于临床医生在临床治疗失败病例中更好地寻找耐药菌株。Objective Using a reverse transcriptase PCR(RT-PCR)-based method to test and optimize the susceptibilities of clinical isolates of urogenital Chlamydia trachomatis in vitro.Methods The 40 clinical strains detected with the method of cultivation by McCoy cells were subcultured until more than 90% McCoy cells were infected.The clinical strains were collected to investigate susceptibilities of doxycycline in vitro.Two methods to determining the minimal inhibitory concentrations(MIC)of doxycycline were the conventional iodine staining and RT-PCR -based method.T test was performed to analyze the difference between the two methods.Results The MICs of doxycycline of the clinical strains were higher by RT-PCR method as compared with iodine staining(P〈0.05).Conclusion The RT-PCR based method may replace the conventional iodine staining method to improve the sensibility of in vitro antibiotic susceptibility of Chlamydia trachomatis.It is helpful for the doctors to find out the drug resistant strains in failed tneatment.
