摘  要:目的分析冷刺激对骨髓脂肪组织的影响,为研究环境对骨质疏松症的影响提供数据。方法选取36只6~8周龄C57BL/6小鼠,采用数字表法随机分为常温对照组和寒冷刺激组。实验结束后取双侧股骨,股骨用于免疫组化、免疫荧光法分析。结果与对照组相比较,冷刺激使骨髓脂肪细胞数目减少(P=0.020),可以提高骨髓脂肪组织中的环磷酸腺苷(cyclic adenosine monophosphate,c AMP)水平(F=23.579,P=0.000),能小幅度提高骨髓脂肪组织中甘油的水平(F=10.086,P=0.003),影响骨髓脂肪组织的脂解率。冷刺激促进小鼠股骨中成骨转录因子Runt相关转录因子2(runt-related transcription factor2,Runx2)的表达,冷刺激对于成脂转录因子过氧化物酶体增生物激活受体γ(peroxisome proliferator activated receptor-gamma,PPARγ)表达的影响不明显。结论冷刺激使骨髓脂肪细胞减少,促进骨髓脂肪组织脂解。Objective To analyze if cold exposure could change the bone marrow adipose tissue,and to provide reliable data for the research between the environment and osteoporosis.Methods A total of 36 C57BL/6 mice (6-8 weeks old) were randomly divided into two groups: the control group and the cold exposure group.After the experiment,the bilateral femur was isolated and analyzed by for immunohistochemistry and immunofluorescence.Results The number of adipose cell in bone marrow adipose tissue was lower in the cold exposure group than those in the control group (P= 0.020).Cold exposure led to an increase of cyclic adenosine monophosphate (cAMP) (F = 23.579,P = 0.000) and glycerol level in bone marrow adipose tissue (F = 10.086,P = 0.003).Cold exposure could promote the expression of runt-related transcription factor2 (Runx2) and inhibit the expression of peroxisome proliferator activated receptor-gamma (PPARγ).Conclusion Cold exposure can reduce the number of adipose cells in bone marrow adipose tissue and promote bone marrow adipose tissue to lipolysis.
