摘  要:用ISSR(inter-simple sequence repeat)技术对60份小叶桦(Betula microphylla Bunge)及变种进行了系统进化分析;从100个引物中筛选出8个可以扩增出多态、重复性好的引物;用该8个引物对全部供试材料的DNA多态性进行检测,共扩增出3 029条谱带,其中多态性条带为2 525条,多态性比率为83.4%。聚类分析结果表明,供试材料相似系数介于0.84~0.90之间,平均相似系数为0.85。取相似系数0.847可将供试材料分为6个类群。小叶桦及其变种在DNA水平上的遗传多样性与形态学特征较一致。该研究为小叶桦类群间的鉴别和优良类群确定及杂交育种等提供了重要的理论依据。Sixty Betula microphylla and their variety samples were analyzed by ISSR (inter-simple sequence repeat) technique to study their taxonomic and phylogenetic relationships. Eight polymorphic and reproducible primers were screened from 100 primers. A total of 3 029 bands were amplified, of which 2 525 were polymorphic bands,and the polymorphic rate was 83.4%. According to ISSR-PCR results, a cluster analysis (UPGMA) was conducted to generate a dendrogram. The results showed that the similarity coefficient of the tested materials were between 0. 84-0. 90, the average similarity coefficient was 0. 85. The test materials were divided into 6 groups at the level of 0. 847. The genetic diversity and morphological characteristics of Betula microphylla and its varieties at the DNA level were more consistent. The study provided an important theoretical basis for the identification of the different taxa and confirmation of excellent taxa and cross breeding in Betula rnicrophylla.
