摘 要:目的通过神经形态学观察FK1706对内脏神经-体神经端侧吻合大鼠内脏神经再生作用。方法30只成年雄性SD大鼠随机分为3组:FK1706组、端侧吻合组和对照组(n=10)。FK1706组:将大鼠左侧腰6前根(L6VR)与腰4前根(L4VR)进行端侧吻合,术后皮下注射FK1706(0.3 mg/kg),连续8周,每周5 d;端侧吻合组:单纯将内脏神经(L6VR)与体神经(L4VR)进行端侧吻合,术后未给予FK1706;对照组:离断左侧腰6后根(L6DR)、骶1前根(S1VR)和骶1后根(S1DR),保持L6VR的完整。术后4个月,取FK1706组和端侧吻合组吻合口远端L6VR延续而成的节前盆副交感神经(PPN),制备半薄切片和超薄切片,计数有髓轴突数量,观察髓鞘形态及厚度并进行比较。 结果术后4个月,FK1706组与端侧吻合组吻合口远端L6VR生长良好,两组大鼠吻合口远端再生PPN有髓轴突数量分别为515.6±43.3和328.4±46.4,两者比较差异有统计学意义(P=0.000)。FK1706组和端侧吻合组大鼠再生PPN的髓鞘厚度分别为(0.672 4±0.110 1) μm和(0.466 8±0.076 3) μm,两者比较差异有统计学意义(P=0.000)。结论FK1706能够提高内脏神经-体神经端侧吻合后内脏神经再生效果,促进外周神经损伤的修复。ObjectiveTo evaluate the effect of FK1706 on nerve regeneration after end-to-side (ETS) neurorrhaphy of autonomic nerve and somatic nerve in rats through morphological observation.MethodsThirty rats were randomly divided into three groups in terms of different treatments (n=10 in each group). In FK1706 group, the left L6 ventral root (L6VR) and L4 ventral root (L4VR) of rats were sutured in an ETS fashion and FK1706 (0.3 mg/kg) was administrated subcutaneously for 5 consecutive days per week for a total of 8 weeks after ETS neurorrhaphy. In ETS group, the same operation as FK1706 group was performed but without FK1706 applied after surgery. In control group, left L6 dorsal root (L6DR), S1 ventral root (S1VR) and S1 dorsal root (S1DR) were transected but the left L6VR was preserved as normal control. After 4 months, the left pelvic parasympathetic nerve (PPN) to which L6VR extended was harvested for semithin and ultrathin sections in FK1706 and ETS groups to count the number of myelinated axons. The number and morphology of myelinated axons and thickness of myelin sheath were compared between the two groups. ResultsAfter 4 months, the morphology of the regenerated L6VR distal to the neurorrhaphy site in FK1706 and ETS groups was normal. The number of regenerated myelinated axons of PPN was 515.6±43.3 and 328.4±46.4 in FK1706 and ETS groups, respectively (P=0.000). The thickness of myelin sheath of regenerated PPN was (0.672 4±0.110 1) μm and (0.466 8±0.076 3) μm in FK1706 and ETS groups, respectively (P=0.000).ConclusionThe effect of autonomic nerve regeneration could be enhanced after ETS neurorrhaphy of autonomic nerve and somatic nerve through FK1706, which could improve rehabilitation of nerve injury.
