摘 要:本研究根据GenBanK发布的核苷酸数据库信息,设计扩增鱼类线粒体COⅢ基因的通用引物,扩增黄渤海地区7种重要商品海鱼(光鱼、梭鱼、鲫鱼、舌鳎、扒皮鱼、六线鱼、白姑鱼)的COⅢ全长基因序列.对扩增产物进行测序、比对,利用分子生物学软件进行序列酶切分析,最终选用Nla Ⅲ限制性内切酶进行酶切,并获得了所研究的各鱼种的特异酶切片段图谱.结果表明,利用聚合酶链式反应-限制性片段长度多态性(PCR-RFLP)技术能有效地区分以上7种海鱼,PCR-RFLP技术应用在鱼种鉴定上具有快速、准确的优势.According to the nucleotide database published in GenBank, a pair of universal primers are designed to amplify a fragment of the mitochondrial COⅢgene in different fish species. The expected fragment of the COⅢgene is amplified from seven important commercial marine fish species in the Yellow Sea and the Bohai Sea, including Acanthogobius hasta , Liza haematocheila , Carassius auratus , Cynoglossus , Thamnaconus modestus , Hexagrammos otakii and Argyrosomus argentatus). The amplified products are sequenced and analyzed with the molecular biology software, NlaⅢ restriction endonuclease is used for digestion, and the specific fragments of the fish species are ob- tained. The results indicate that polymerase chain reaction-based restriction fragment length polymorphism (PCR- RFLP) can effectively distinguish the above seven marine fish species. PCR-RFLP has the advantages of fast and accurate in fish species identification.
