摘  要:目的研究养阴清热、润肺止咳验方防治放射性肺损伤的作用机制。方法用射线方法建立放射性肺损伤大鼠模型。将60只模型大鼠随机分为阳性对照组、模型组、实验A组、实验B组、实验C组,每组12只;另取,24只健康大鼠随机分为空白组和阴性对照组,每组12只。照射完成后次日,实验A、B、C组分别予以1,2,4 g·m L^(-1)养阴清热、润肺止咳验方;阳性对照组予以地塞米松;模型组和空白组均予以蒸馏水;阴性对照组予以2 g·mL^(-1)养阴清热、润肺止咳验方。7组大鼠均按0.01 m L·g^(-1)的剂量,每天灌服一次,总共14 d。14 d后处死大鼠,用Western blot法检测肺组织细胞中髓样分化蛋白88(My D88)、肿瘤坏死因子受体活化因子-6(TRAF-6)、诱导干扰素β的TIR基域接头分子(TRIF)、Toll样受体4(TLR4)的表达量,用酶联免疫吸附法检测肺组织匀浆上清液中肿瘤坏死因子-α(TNF-α)、白细胞介素-17(IL-17)、转化生长因子β(TGF-β)表达量。结果实验A、B、C组、模型组、阳性对照组、阴性对照组及空白组的My D88的灰度值(×10^(-2)))分别为(60.04±6.95),(51.75±6.50),(53.31±3.80),(63.27±0.06),(53.29±2.23),(21.43±5.42)和(21.16±4.81),TRAF-6的灰度值(×10^(-2)))分别为(77.44±4.16),(70.26±6.48),(66.05±4.17),(80.58±3.82),(65.23±2.60),(56.31±7.40)和(54.51±7.81),TRIF的灰度值(×10^(-2))分别为(48.01±6.51),(40.80±5.16),(41.20±5.18),(48.62±4.10,(39.44±4.25),(33.83±3.40和(32.49±4.71),TLR4的灰度值(×10^(-2)))分别为(65.13±4.53),(59.46±3.59),(57.13±4.83),(67.45±3.73),(56.65±3.82),(36.52±3.50)和(40.19±3.07),TNF-α分别为(255.71±4.87),(215.77±2.83),(211.27±3.81),(283.86±8.30),(208.21±1.31),(87.15±3.60)和(105.55±1.81)pg·m L^(-1),IL-17分别为(45.56±2.66),(32.84±3.64),(32.66±1.78,(48.56±1.26),(28.74±1.65),(17.26±3.00)和(20.44±2.69)pg·m L^(-1),TGF-β分别为(64.24±3.49),(53.19±4.30),(52.34±6.49),(67.09±6.04),(46.76±2.93),(38.22±6.16)和(38.37±1.74)ngObjective To explore the mechanism of Yangyinqingre andRunfeizhike experiential prescription in the treatment of radiation induced lung injury. Methods The model of radioactive lung injure rats was established by radiation. Sixty model rats were randomly divided into positive control group,model group,Test A group,Test B group,Test C group with 12 rats per group. Another 24 health rats were randomly divided into blank group and negative control group with 12 rats per group. At the next day after radiation,test A,B,C groups were given 1,2,4 g·m L^-1angyinqingre and Runfeizhike experiential prescription,positive control group was given dexamethasone,model and blank groups were given water,negative control group was given 2 g·m L^-1Yangyinqingre and Runfeizhike experiential prescription. The dose was 0. 01 m L·g^-1for 7 groups with qd for 14 days. after 14 d the rats were sacrificed,the expression of Myeloid differentiation factor 88( My D88),tumor necrosis factor receptor associated factor-6( TRAF-6) and TIR domain containing adaptor inducing( TRIF) and Toll-like receptor 4( TLR4) were measured by Western blot method,the expression of tumor necrosis factor-α( TNF-α),interleukin-17( IL-17) and transforming growth factor beta( TGF-β) were measured by enzyme-linked immunosorbent assay( ELISA).
